口腔鳞癌中hTRT mRNA的表达及其与PCNA关系的探讨 被引量:4

摘要 目的 :探讨端粒酶逆转录酶 (hTRT)在口腔鳞癌、癌旁组织的表达及其与增殖细胞核抗原 (PCNA)之间的关系。方法 :用原位杂交技术及免疫组化方法检测 5 2例口腔鳞癌标本。结果 :正常癌旁上皮、上皮异常增生组织及鳞癌hTRTmRNA阳性率分别为 2 0 %、41.7%、82 .7%。PCNA阳性组与阴性组间hTRT表达水平有显著差异 (P <0 .0 5 )。结论 :hTRT在口腔鳞癌发生、发展中起重要作用 ,hTRT与PCNA阳性表达呈正相关 ,表明hTRT激活细胞多处于高增殖状态。 砄bjective: To investigate Human telomerase reverse transcriptase(hTRT) mRNA expression in oral squamous cell carcinoma(OSCC) and the relationship between hTRT activity and proliferation cell nuclear antigen(PCNA) expression.Methods: The expression of hTRT mRNA and PCNA was detected by in situ mRNA hybridization and immuno histochemical technique in 52 cases of OSCC and 22 cases of OSCC adjacent tissues including histologically normal mucosas and dysplastic lesions. Results: hTRT mRNA was observed in 82.7% cases of OSCC, 41.7% of dysplasia and 20% of normal mucosa( P <0.05). Among 36 PCNA positive cases, 33 of them were hTRT mRNA positive, while 3 negative; among 16 PCNA negative cases 10 of them were hTRT mRNA positive, while 6 negative ( P< 0.05). Conclusion: Telomerase subunit hTRT may play a crucial role in the tumorigenesis and development of OSCC, especially in early stage.
