摘要
检测了宫颈鳞癌患者放疗前和放疗中凋亡指数 (AI)及bcl- 2、bax的基因表达 ,探讨了宫颈癌组织中基因表达与凋亡之间的关系。采用免疫组化SABC法检测bcl- 2和bax基因的表达 ,TUNEL法检测宫颈癌活检组织的细胞凋亡水平。在此基础上分析bcl- 2和bax基因表达的细胞阳性率与凋亡指数的关系。结果表明 ,X射线照射 10Gy后 ,宫颈癌组织bax基因表达增强 (p<0 .0 5 ) ,凋亡指数亦明显增加 (p <0 .0 1) ,bcl- 2基因表达的细胞阳性率水平无明显改变 (p >0 .0 5 )。照射前宫颈癌组织bcl- 2和bax的基因综合表达细胞阳性率与自发凋亡相关性较差 (p >0 .0 5 ,r=0 .370 3,其上界为 0 .70 71) ,而 10Gy照射后 ,宫颈癌组织中bcl- 2和bax的综合表达细胞阳性率与辐射诱导的凋亡具有明显的相关性 (p <0 .0 5 ,r =0 .5 2 6 4,其上界为 0 .70 71)。
The aim of this paper was to observe bcl-2 and bax gene expression and apoptosis and to study the correlation between bcl-2 and bax gene expression and apoptotic index in the samples from patients with cervical squamous cell carcinoma following radiotherapy. Apoptosis was detected with TUNEL method. It was found that the bax gene expression and apoptotic index in cervical carcinoma significantly increased ( p <0.05 and p <0.01, respectively) after 10Gy irradiation compared with those of pre-therapy. However, the bcl-2 gene expression had no obvious change ( p >0.05). Our data suggested that the bcl-2 and bax gene cooperative expression was related unobviously with spontaneous apoptosis, but obviously with radiation-induced apoptosis( p <0.05, r =0.5264) in patients with cervical squamous cell carcinoma.
