摘要
采用(NH4)2SO4 分段盐析和葡聚糖凝胶SephadexG 100柱层析纯化技术 ,从九孔鲍Haliotisdiversicolorsupertexta内脏器官中提取纯化褐藻酸酶、琼脂酶及纤维素酶。结果表明 ,在(NH4)2SO4分段盐析纯化中 ,褐藻酸酶和纤维素酶的最适分离饱和度为60% ,而琼脂酶为70%。分段盐析的提纯倍数为(以粗酶提取液为参照)褐藻酸酶13.3 ,琼脂酶8.7和纤维素酶10.9。葡聚糖凝胶SephadexG 100层析分离过程中 ,褐藻酸酶、琼脂酶和纤维素酶的比活力高峰分别出现在洗脱液的64,48和80ml处 ,提纯倍数分别为褐藻酸酶80.9 ,琼脂酶68.0及纤维素酶15.2。
Algalase,agarase, and cellulase were extracted and purified from the abalone Haloitis diversicolor supertexta by the means of (NH 4)2SO 4 fractinated salting out and gel filtration Sephadex G 100. In the means of (NH 4)2SO 4 fratonated salting out, the 60% is the optimum saturation degree for the enzymes of algalase and cellulase, as well as the 70% is the optimum for the enzyme of agarase. The number of purification times of algalase was 13.3 (crude extraction liquid as control ),8.7 for agaraseand 10.9 for cellulase. The specific activity peaks were showed out in the washing off liquid of 64 ml for algalase,48 ml for agarase and 80 ml for cellulase, as well as the number of the purification times were 80.9 for algalase, 68.0 for agarase and 15.2 for cellulase. The above study results on the three enzymes can be used to study the specifications and to make the enzyme preparations afterward.
