摘要
目的 探讨生产工人吸烟、被动吸烟对淋巴细胞DNA损伤的影响。方法 用彗星电泳方法测定 35 9名电梯厂工人 (男性 2 5 2人 ,女性 10 7人 )外周血淋巴细胞DNA的损伤 ,2 0 5人为生产作业工人 ,另 15 4人为管理人员 (对照组 )。结果 吸烟可使DNA损伤增加 (P <0 0 0 1) ,测定的彗星矩 (TailMoment ,TM)以每日吸烟者为最大 ( 0 93μm) ,其次为偶尔吸烟者 ( 0 76μm)和戒烟者 ( 0 70 μm) ,而以从不吸烟者为最小 ( 0 5 6μm) ;被动吸烟亦可使TM增加 (P =0 0 2 1) ;生产工人DNA损伤增加的TM值远大于管理人员 [0 74μm( 0 68~ 0 80 )与 0 5 3μm( 0 49~ 0 5 6) ,P <0 0 0 1] ;协方差分析表明吸烟 (P <0 0 0 1)、被动吸烟 (P <0 0 32 )和性别 (男性 ,P <0 0 0 1)对TM值有影响 ,DNA损伤与年龄 (P =0 0 0 1)呈正相关。结论 吸烟、被动吸烟可致淋巴细胞DNA损伤 ,男性较女性DNA损伤为重 。
Objective This study aimed at studying the effects of smoking, passive smoking, alcohol drinking and occupational exposure on DNA strand breaks in elevator manufacturing workers.Methods Of 359 workers (252 men and 107 women), 205 were from production departments and 154 from management department. DNA damages were measured by the comet assay. Results The tail moment (TM) appeared to be significantly increased by smoking ( P <0.001). Daily smokers had the largest TM [Geometric mean, 95% CI ,0.93 μm(0.81-0.94)], followed by occasional smokers, ex smokers, and never smokers. Passive smoking also caused the significant increase of the TM ( P =0 021). Production workers had a larger TM than management workers did(0 74 μm vs 0 53 μm, P <0 001). Men had a larger TM than women did(0 70 μm vs 0 51 μm, P <0 001).TM was shown to be significantly increased with age after adjustment for occupational exposure, smoking, passive smoking and drinking (partial r=0 17,P= 0 001). Analysis of covariance showed that smoking ( P <0 001),passive smoking ( P =0 031), gender ( P <0 001) and age ( P =0 001) had independent effect on tail moment. Conclusion Smoking, passive smoking and occupational exposure might independently increase the DNA strand breaks. Age was linearly correlated with TM ( P =0 001).
