基于整合药理学分析通脉养心丸治疗冠心病心绞痛及心律不齐的分子机制 被引量:9

Molecular mechanism of Tongmai Yangxin Pills for angina and arrhythmia of the coronary heart disease based on integrative pharmacology

摘要 目的:预测通脉养心丸治疗冠心病心绞痛及心律不齐的分子机制,为通脉养心丸有效成分辨识、分子机制与临床应用提供科学依据。方法:通过整合药理学平台,进行通脉养心丸化学成分分析、疾病靶标预测、基因功能和通路富集分析,并建立"中药方剂-化学成分-作用靶标-疾病靶标"多维度关联的网络关系,对通脉养心丸治疗冠心病心绞痛及心律不齐的作用机制进行探讨。结果:通脉养心丸药物靶标与冠心病心绞痛及心律不齐疾病靶标基因功能分析结果显示有细胞质、胞外谷氨酸门控离子通道活性、离子型谷氨酸受体信号通路、线粒体、ATP结合、用于蛋白质翻译的tRNA氨酰化等条目。通路富集分析结果显示主要集中在谷氨酸能突触、神经系统、氨基酸代谢、丙氨酸,天冬氨酸和谷氨酸代谢、精氨酸和脯氨酸代谢等信号通路。通脉养心丸作用于冠心病心绞痛心律不齐的中药中,生地黄、党参、阿胶、大枣的化学成分及预测靶标数居多。结论:通脉养心丸通过离子通道、线粒体途径、ATP能量代谢、神经系统等相关通路来达到对冠心病心绞痛及心律不齐的治疗作用。本研究还得到通脉养心丸发挥这一作用的主要药物可能是生地黄、党参、阿胶和大枣。 Objective:To find the molecular mechanism of Tongmai Yangxin Pills in treating angina and arrhythmia of the coronary heart disease,to provide scientific basis for identification of effective ingredients,molecular mechanism and clinical application of Tongmai Yangxin Pills.Methods:In this study,we used the internet-based Computation Platform (www.tcmip.cn) to analyze chemical composition,predict disease target,analyze gene function and pathway,and establish a multi-dimensional network relationship of 'Chinese medicine formula-chemical composition-candidate target -disease target'.Results:The results of gene function analysis of Tongmai Yangxin Pills target and Angina and arrhythmia of the coronary heart disease target showed cytosol,extracellular-glutamate-gated ion channel activity,ionotropic glutamate receptor signaling pathway,mitochondrion,ATP binding,tRNA aminoacylation for protein translation and other items.Pathway enrichment analysis results display glutamatergic synapse,nervous system,amino acid metabolism,alanine,aspartate and glutamate metabolism,arginine and proline metabolism, etc.TMYXP was used for Angina and arrhythrnia of the coronary heart disease,Rehmannia glutinosa libosch,Codonopsis radix, Asini Corii Colla,Fructus Jujubae have the majority of chemical components.Conclusion:Tongmai Yangxin Pills is used for Angina and arrhythmia of the coronary heart disease through ion channel,mitochondrial pathway,ATP energy metabolism,and nervous system related pathways.Rehmannia glutinosa libosch,Codonopsis radix,Asini Corii Colla,Fruetus Jujubae may have the majority of chemical components in Tongmai Yangxin Pills.
