红景天苷减轻手机电磁辐射对大鼠睾丸支持细胞损伤的实验研究 被引量:4

摘要 [目的]探究红景天苷在手机电磁辐射诱导睾丸支持细胞损伤中的保护作用。[方法]体外分离培养大鼠睾丸支持细胞,并分为5组,对照组、辐射组、辐射+低剂量红景天苷组、辐射+中剂量红景天苷组和辐射+高剂量红景天苷组。对照组不作任何处理,其余组均接受频率为900MHz的电磁辐射,24h/d,辐射+低剂量红景天苷组、辐射+中剂量红景天苷组和辐射+高剂量红景天苷组同时分别加入终浓度分别为100、200、400μg·mL-1的红景天苷,继续培养5d后,CCK-8法检测各组细胞增殖活性;紫外分光光度法检测细胞培养上清液中超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)、谷胱甘肽过氧化酶(glutathione peroxidase,GPx)活性及丙二醛(malondialdehyde,MDA)含量;FQ-PCR检测各组细胞雄激素结合蛋白(androgen binding protein,ABP)、抑制素βB亚基(inhibinβB,INHβB)mRNA表达水平。[结果]各组细胞增殖活性差异有统计学意义(F=153.93,P<0.01);与对照组比较,辐射组细胞增殖活性明显降低(P<0.01);与辐射组比较,辐射+中剂量红景天苷组和辐射+高剂量红景天苷组细胞增殖活性增高(P<0.01)。各组间SOD、CAT、GPx活性和MDA含量差异均有统计学意义(P<0.01);与对照组比较,辐射组SOD、CAT和GPX活性均明显降低,MDA含量明显增加(P<0.01);与辐射组比较,辐射+中剂量红景天苷组和辐射+高剂量红景天苷组SOD、CAT和GPX活性逐渐升高,MDA含量逐渐下降(P<0.05或P<0.01)。各组细胞中ABP和INHβB mRNA表达量差异均有统计学意义(P<0.01);与对照组比较,辐射组ABP和INHβB mRNA表达均明显降低(P<0.01);与辐射组比较,辐射+中剂量红景天苷组和辐射+高剂量红景天苷组ABP和INHβB m RNA表达均增高(P<0.05或P<0.01)。[结论]红景天苷可降低手机电磁辐射对睾丸支持细胞分泌ABP和INHβB的影响,减轻睾丸支持细胞氧化损伤。 [Objective]To explore the effect of salidroside in sertoli cells injury induced by cellphone electromagnetic radiation.[Methods]The rats sertoli cells were isolated in vitro and primarily cultured,and then grouping it with control group,radiated group and radiated +three different dose salidroside groups.Control group didn't do any treatment,other groups exposed electromagnetic radiation of 900MHz cellphone for 5days.Meanwhile,The final concentration 100μg·mL^-1,200μg·mL^-1,400μg·mL^-1 salidroside were added into radiated +low dose salidroside,group radiated +middle dose salidroside group and radiated +high dose salidroside group.The cell proliferation was detected by CCK-8;superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GPx)activity and malondialdehyde(MDA)concentration were detected by ultraviolet spectrophotometry;the relative expression quantity of androgen binding protein(ABP)and inhibinβB(INHβB)mRNA were detected by FQ-PCR.[Results]The difference of cells proliferation in all groups was obvious(P<0.01).Compared with control group,cell proliferation of radiated group was reduced(P<0.01),and compared with radiated group,these of radiated +middle dose salidroside group and radiated +high dose salidroside group were raised(P<0.01).The difference of SOD,CAT,GPx activity and MDA concentration in groups was obvious(P<0.01,respective).Compared with control group,radiated group SOD,CAT and GPx activity were reduced(P< 0.01,respective),MDA concentration raised(P<0.01).Compared with radiated group,SOD,CAT,GSH-Px activity in radiated+middle and high dose salidroside groups gradual raised,and MDA concentration gradual reduced(P<0.05or P<0.01).The difference of ABP and INHβB mRNA relative expression quantity in four groups wele obvious(P<0.01,respective).Compared with control group,the ABP and INHβB mRNA relative expression quantity of radiated group were decreased(P<0.01),and compared with radiated group,ABP and INHβB mRNA expression in radiated+middle and high dose salidroside group's increased(P<0.05or P<0.01).[Conclusion]Salidroside could reduce oxidative damage induced by eelIphone electromagnetic radiation,and promoting ABP and INHβB expression in sertoli ceils in vitro.
