摘要
目的:应用SSR标记研究海拔对贵州雷公山地区南方红豆杉遗传多样性的影响。方法:经梯度PCR扩增从红豆杉、白云杉、日本柳杉SSR引物中筛选获得了条带清晰的21对SSR引物用于群体研究。结果:21对引物共扩增出42条条带,其中多态性条带35条,多态性条带百分率为83.3%;物种水平上Nei's基因多样性为0.2587,Shannon指数为0.4719,多态位点百分率为85.71%,基因流为1.1453。聚类分析显示在阈值为0.71时,不同海拔南方红豆杉种群之间出现遗传分化。结论:海拔高度与南方红豆杉种群的遗传分化呈负相关。
Objective:The effects of altitude on genetic diversity for Taxus chinensis var.mairei at Leigongshan area were studied by using the SSR Molecular marker technology.Methods:21 groups of SSR primers were selected from SSR primers in Taxus cuspidata, Picea glauca and Cryptomeria japonica by using gradient PCR amplification and used to study the samples.Resuhs:A total of 42 bands were amplified by 21 pairs of primers ,which 35 bands were polymorphic and the percentage of polymorphic bands was 83.3%.At the species level,the Nei's gene diversity was 0.2587,the Shannon index was 0.4719,the percentage of palymorphic loci was 85.71%,and the gene flow was 1.1453.Cluster analysis showed that there was the genetic differentiation between Taxus chinensis vat.mairei populations at different altitudes when the threshold was 0.71.Conclusion:There is a negative correlation between the altitude and genetic differentiation in Taxus chinensis vat.mairei populations.
