摘要
目的:观察赤芍总苷与白芍总苷对给予不同预收缩刺激的大鼠肠系膜上动脉的舒张作用。方法:高效液相检测白芍总苷和赤芍总苷中主要有效成分比例;采用DMT Myograph肌动描计系统记录离体血管张力。观察赤芍总苷与白芍总苷(10^(-6)~10-2g/L)对基础状态和用K+(60mM)、PE(10^(-5)M)、5-HT(10^(-5)M)分别预收缩的内皮完整血管的舒张作用;观察赤芍总苷与白芍总苷(10^(-6)~10-2g/L)对用K+(60mM)、PE(10^(-5)M)、5-HT(10^(-5)M)分别预收缩的去内皮血管环的舒张作用。结果:赤芍总苷中主要有效组分氧化芍药苷、芍药内酯苷和芍药苷含量分别为24.72%、19.66%、41.24%;白芍总苷中主要有效组分氧化芍药苷、芍药内酯苷和芍药苷含量分别为5.54%、21.38%、11.78%;在内皮完整的血管上,赤芍总苷与白芍总苷对基础状态的血管没有舒张作用;在不同物质刺激预收缩的内皮完整及去内皮的血管上,赤芍总苷在(10^(-4)~10-3g/L)剂量区间范围内对用K+、PE及5-HT预收缩的血管均有显著舒张作用,而白芍总苷的舒血管作用不明显。结论:用同样工艺流程提取的赤芍总苷和白芍总苷其舒血管作用有明显差别,可能与其有效组分芍药苷、氧化芍药苷和芍药内酯苷等物质的含量差别有关。
Objective:To investigate the vasodilative effects of Total Glucosides from Red Paeony(TPG) and White Paeony Total Glycoside(TGP)on rat superior mesenteric artery in vitro.Methods:HPLC was applied to determine contents of TGP and TPG with Diamonsil C18 column.By using DMT Myograph in vitro vascular tension meter system,we assessed the muscular responses of rat superior mesenteric arterial ring to TGP and TPG.Also we observed the diastolic function of the intact or denuded endothelial rats’ superior mesenteric artery in basis state and stimulus state by K+(60 mM),PE(10-5 M) and 5-HT(10-5 M),respectively.Results:The main components in the TGP were oxypaeoniflorin(5.54%),albiflorin std(21.38%),paeoniflorin(11.78%).In TPG the main components were oxypaeoniflorin(24.72%),albiflorin std(19.66%),paeoniflorin(41.24%).TGP and TPG showed no vasodilative effect on intact endothelial artery,but significantly vasodilatation on TPG was observed on K+,PE and 5-HT pre-stimulated intact artery or denuded artery,at the dose of 10-4~ 10-3 g/L(P<0.05,P<0.01),while no significant effect was found for TGP.Conclusion:Extracted through the same process,the significant difference of vasodilatation between TGP and TPG may be related to their effective components of oxypaeoniflorin,albiflorin std and paeoniflorin.
