背部循经推拿对亚健康模型大鼠HPA轴及下丘脑CRH蛋白表达的影响 被引量:9

Effect of Chinese Massaging Along Meridians on Back on HPA Axis and Expression of CRH Protein in Hypothalamus of Sub-health Model Rats

摘要 目的:观察背部循经推拿手法对慢性束缚应激亚健康模型大鼠下丘脑促肾上腺皮质激素释放激素(CRH)mRNA与CRH蛋白表达及血清促肾上腺皮质激素(ACTH)、皮质酮(CORT)的影响,探讨推拿调理亚健康的作用机制。方法:将60只wistar成年大鼠随机分为3组:正常组、模型组与推拿干预组,每组20只。模型组连续21 d造模干预,每天给予3 h的连续束缚以限制其活动。干预组于每日造模后1 h内给予背部循经推拿干预。正常组正常饲养,每天亦经过前两组同样的抓取、固定而非束缚等过程。造模干预结束后采用实时-荧光定量PCR (RT-PCR)方法检测下丘脑CRH mRNA含量,Western Blot法测CRH蛋白表达,酶联免疫吸附法(ELISA)检测大鼠血清ACTH与CORT。结果:(1)与正常组比较,模型组大鼠下丘脑CRH mRNA及其蛋白表达量升高(P<0.05);与模型组比较,干预组大鼠下丘脑CRH mRNA及其蛋白表达量均明显下降(P<0.05)。(2)模型组血清CORT浓度高于正常组(P<0.05),干预组CORT浓度较模型组低(P<0.05),与正常组比较差异无显著性(P>0.05);模型组、干预组血清ACTH浓度均高于正常组(P<0.05),但干预组ACTH浓度低于模型组,差异有显著性(P<0.05)。结论:慢性应激致亚健康模型大鼠出现下丘脑-垂体-肾上腺皮质(HPA)轴功能亢进,推拿干预能够下调下丘脑的CRH mRNA表达从而下调血清CORT与ACTH含量,达到拮抗亚健康状态下HPA轴的功能亢进,改善亚健康状态身心症状,这是推拿调理亚健康的作用机制之一。 Objective:To observe the effect of back massage on the hypothalamus of chronic restraint stress sub-health model rats’ corticotropin releasing hormone(CRH) mRNA and CRH protein expressions and the influence on serum adrenocorticotropic hormone(ACTH) and corticosterone(CORT) and discuss massage regulating sub health mechanism. Methods:Sixty Wistar adult rats were randomly divided into 3 groups: normal group,model group and massage intervention group,with 20 rats in each group. The model group was given 21 days of model intervention,and 3 hours of continuous restraint was given daily to limit its activity. The intervention group was given the back-through massage intervention within 1 h after a daily model. The intervention group was given an intervention of the back in 1 h after daily modeling. Normal group was given normal feeding and the same grasping. The CRH mRNA content in the hypothalamus was detected by real-time fluorescence quantitative PCR(RT-PCR), and the expression of CRH protein was detected by Western blot,and serum ACTH and CORT were detected by enzyme-linked immunosorbent assay(ELISA). Results:(1)Compared with the normal group, the expression of CRH mRNA and protein in the hypothalamus of the model group increased(P<0.05). Compared with the model group,the expression level of CRH mRNA and its protein in hypothalamus of intervention group decreased significantly(P<0.05).(2)The model group’s serum CORT concentration was higher than that of the normal group(P<0.05). The intervention group’s CORT concentration was lower than that of the model group(P<0.05). Compared with the normal group,there was no significant difference(P>0.05). Model group and intervention group’s serum ACTH concentrations were higher than that of the normal group(P<0.05), but the intervention group’s ACTH was lower than that of the model group and there was significant difference(P<0.05). Conclusion:Chronic stress caused by sub-health model rats appeared the hypothalamus-pituitary-adrenal(HPA)axis hyperfunction. The massage intervention can down regulate the expressions of mRNA and CRH in the hypothalamus,decrease serum CORT and ACTH contents to reach hyperfunction of HPA axis antagonist sub-health condition, improve physical and mental symptoms of sub-health state,which is one of the mechanisms of massage to regulate sub-health.
