基于网络药理学浅析牡丹叶治疗宫颈癌的分子机制 被引量:7

Network pharmacology based study on mechanism of peony leaf in the treatment of cervical cancer

摘要 目的基于网络药理学浅析牡丹叶治疗宫颈癌的多成分、多靶点、多途径作用机制。方法通过中药系统药理学数据库和分析平台对牡丹叶可能的入血活性成分和靶点进行筛选和预测,通过搜索Therapeutic Target Database等数据库得到宫颈癌相关靶点,然后针对牡丹叶入血活性成分筛选出其调控疾病的潜在靶点,再根据网络拓扑特征参数值筛选出牡丹叶治疗疾病的关键靶点名称及其所参与的通路,并通过ClueGO加以富集分析。结果牡丹叶11个入血成分的作用靶点约为194个,最后共得到显著富集的信号通路171条,排除宽泛通路后共得到与宫颈癌相关关键通路21条,如雌激素信号通路、神经营养素信号通路等。结论牡丹叶可能通过作用于炎症、代谢、免疫、内分泌及细胞周期等相关的蛋白靶点和通路起到治疗宫颈癌的作用。 ObjectiveTo analyze the multicomponent, multi-target and multipathway mechanism of peony leaf in the treatment of cervical cancer based on network pharmacology. MethodsThe possible active components and targets in peony leaf were screened and predicted by pharmacological database and analysis platform of traditional Chinese medicine system, and the related targets of cervical cancer diseases were obtained by searching Therapeutic Target Database and others. The potential targets for the disease regulation were screened according to the active components of peony leaves entering blood, then the key target names and the pathways involved in the treatment of peony leaf were selected according to the network topological characteristic parameters. Then, the enrichment analysis was carried out by using ClueGO software plugin. ResultsThere were 194 target sites for 11 blood entry components in peony leaves. Finally, 171 signal pathways were obtained, and 21 key pathways related to cervical cancer were obtained after the wide pathway was excluded, such as estrogen signaling pathway, neurotrophin signaling pathway and so on. ConclusionPeony leaves may play a vital role in the treatment of cervical cancer by acting on inflammatory, metabolic, immunological, endocrine and cell cycle related protein targets and pathways.
