活血解毒方对NOD小鼠LincR-Ccr2-5’AS、CCR5及CCL5表达的影响 被引量:4

Effect of Huoxue Jiedu Formula on the expression of LincR-Ccr2-5'AS,CCR5 and CCL5 in NOD mice

摘要 目的观察活血解毒方对干燥综合征(SS) NOD小鼠颌下腺LincR-Ccr2-5’AS、趋化因子受体5(CCR5)及趋化因子配体5(CCL5)表达的影响。方法 34只NOD/Ltj小鼠随机分为模型组、对照组、中药组,12只C57BL/6J小鼠作为正常组。正常组、模型组按10 m L/kg灌服蒸馏水,对照组按234 mg/kg灌服白芍总苷,中药组按28 g/kg灌服活血解毒方,共干预8周。ELISA法检测血清CCL5水平; HE染色颌下腺组织,镜下观察其形态学变化并评分; RT-qPCR法检测颌下腺组织中LincR-Ccr2-5’AS、CCR5 mRNA、CCL5 mRNA表达。结果模型组血清CCL5水平高于正常组(P <0. 01),对照组、中药组血清CCL5水平均低于模型组(P <0. 01),中药组血清CCL5水平低于对照组(P <0. 01)。正常组组织学评分低于各组NOD小鼠(P <0. 01),各组NOD小鼠组间组织学评分差异无统计学意义(P> 0. 05)。中药组颌下腺LincR-Ccr2-5’AS表达低于模型组(P <0. 01),对照组颌下腺CCL5 mRNA表达低于模型组(P <0. 01),各组颌下腺CCR5 mRNA表达差异无统计学意义(P>0. 05)。颌下腺组织中CCR5 mRNA表达与CCL5 mRNA表达呈正相关(r=0. 512,P <0. 01),CCR5 mRNA与血清CCL5水平呈正相关(r=0. 413,P <0. 05),CCL5 mRNA与血清CCL5呈正相关(r=0. 377,P <0. 05)。结论活血解毒方可下调NOD小鼠血清CCL5水平及颌下腺组织中LincR-Ccr2-5’AS表达,对颌下腺组织中CCR5及CCL5表达无明显影响。 Objective To observe the effect of Huoxue Jiedu Formula on the expression of LincR-Ccr2-5'AS in submandibular gland of non-obese diabetic (NOD)mice with Sjogren's syndrome(SS).Methods 34NOD/Ltj mice were randomly divided into model group,control group and Chinese medicine group,and 12C57BL/6J mice were used as normal group.The normal group and the model group were given distilled water at 10mL/kg.The control group was given total glucosides of paeony at 234mg/kg.The Chinese medicine group was treated with Huoxue Jiedu Formula at 28g/kg,all for 8weeks.The eyeballs of mice of each group were taken for blood and serum CCI.5levels were detected by ELISA.The submandibular gland tissues were stained by HE,and observed under the microscope and scored.The expressions of LincR-Ccr2-5'AS,CCRSmRNA and CCL5mRNA in the submandibular gland were detected by RT-qPCR.Results Serum CCI-5in the model group was higher than that in the normal group(P <0.01).The serum CCL5level of the control group and Chinese medicine group was lower than that of the model group(P <0.01).The serum CCL5level of Chinese medicine group was lower than that of the control group(P <0.01).The histological scores of the normal group were lower than those of NOD mice of other groups(P <0.01).There was no significant difference in the histological scores between the groups of NOD mice (P >0.05).The expression of LincR-Ccr2-5'AS in the submandibular gland of Chinese medicine group was lower than that of the model group(P <0.01).The expression of CCL5mRNA in the submandibular gland of the control group was lower than that of the model group(P <0.01).There was no significant difference in the expression of CCR5mRNA in the submandibular glands between the two groups(P >0.05).There was a positive correlation between CCR5mRNA expression and CCL5mRNA expression in submandibular gland tissue(r =0.512,P <0.01).CCR5mRNA was positively correlated with serum CCL5level(r =0.413, P <0.05).CCI.5mRNA was positively correlated with serum CCL5(r =0.377,P <0.05).Conclusion Huoxue Jiedu Formula can down-regulate the serum CCL5level in NOD mice and the expression of LincR-Ccr2-5'AS in submandibular gland tissue,and has no significant effect on the expression of CCR5and CCL5in submandibular gland.
