MiR-33a在胃癌组织中低表达并抑制HGC-27细胞增殖、迁移与侵袭 被引量:1

MiR-33a is poorly expressed in gastric cancer tissues and inhibits the proliferation, migration and invasion of HGC-27 cells

摘要 目的探讨miR-33a在胃癌患者中的表达,探讨miR-33a对胃癌HGC-27细胞增殖、侵袭和转移的影响。方法收集胃癌患者标本39例,同时收取相应的癌旁组织20例。通过荧光定量PCR方法检测各组织中miR-33a的表达水平。以脂质体为载体将miR-33a模拟物转染至HGC-27细胞,CCK-8方法检测细胞增殖能力变化,Transwell实验检测HGC-27细胞侵袭和迁移能力变化。结果 miR-33a在胃癌组织中的表达水平明显低于癌旁组织,过表达miR-33a能够显著抑制HGC-27细胞的增殖,显著抑制HGC-27细胞的侵袭和迁移能力。结论 miR-33a可能对胃癌有治疗作用。 Objective To investigate the expression of miR-33 a in gastric cancer patients and explore the effect of miR-33 a on the proliferation, invasion and metastasis of gastric cancer HGC-27 cells. Methods The expression of miR-33 a was detected by fluorescence quantitative PCR in 39 patients with gastric cancer and 20 corresponding para-cancerous tissues. The miR-33 a mimics were transfected into HGC-27 cells with liposome, and cell proliferation was detected by CCK-8, the invasion and migration ability of HGC-27 cells were detected by transwell test. Results The expression level of miR-33 a was significantly lower in gastric cancer tissues than in adjacent normal tissues. The over-expression of miR-33 a could significantly inhibit the proliferation, invasion and migration of HGC-27 cells. Conclusion miR-33 a might be usefull for the teatment of gastric cancer cells.
