摘要
目的分析益气活血中药组分对慢性心力衰竭实验大鼠血清NT-proBNP和cTnI水平的影响研究。方法选取80只健康的SD大鼠,随机选取20只作为空白组,其余60只大鼠通过左冠脉动脉前降支结扎法加力竭游泳法建模,将建模成功的大鼠随机分为建模组、益气活血组和赖诺普利组。空白组与建模组给予等容积蒸馏水灌胃,益气活血组给予益气活血中药配方制剂灌胃,赖诺普利组给予赖诺普利灌胃。在持续给药28天后大鼠禁食12 h后取血清。利用ELISA方法检测NT-proBNP,及利用电化学发光法检测cTnI水平。结果与空白组相比,建模组、益气活血组、赖诺普利组的NT-proBNP和心cTnI水平均升高,组间差异具有统计学意义(P <0.05);与建模组比较,益气活血组、赖诺普利组的NT-proBNP和cTnI水平均降低,且差异具有统计学意义(P <0.05);而益气活血组和赖诺普利组组间无明显统计差异(P> 0.05)。结论益气活血中药能明显降低慢性心衰模型大鼠血清NT-proBNP及cTnI水平,通过减轻心室重构的过程达到治疗慢性心力衰竭的目的。
Objective To measure the components of the Chinese materia medica for benefiting qi for activating blood circulation on chronic heart failure (CHF) rats after myocardial infarction. Methods Twenty rats were selected from eighty healthy SD rats as the blank control group, and the rest were used to prepare CHF rat model by coronary artery ligation in combination with exhausted swimming and food reducing. Successful modal-building rats were randomly divided into model group, Chinese materia medica group and Lisinopril group. Blank control group and model group were given conventional volume distilled water to fill the stomach, the Chinese materia medica group were given the preparation of Chinese materia medica for benefiting qi for activating blood circulation to fill the stomach, Lisinopril group were given Lisinopril to fill the stomach. After sustained dosing of 28 days, fast and only drink water for 12 hours, rats were sacrificed to get myocardial tissue and plasma. The ELISA assay was conducted to measure NT-proBNP level, and electrochemiluminescence immunoassay was used to test cTnI level. Results Compared with the blank control group, the NT-proBNP and cTnI level were increased in modal group, Chinese materia medica group and Lisinopril group, showing statistical difference ( P < 0.05). Compared with the model group, the NT-proBNP and cTnI were decreased in Chinese materia medica group and Lisinopril group, showing statistical difference ( P <0.05). There was no significant difference of NT-proBNP and cTnI level between Chinese materia medica group and Lisinopril group ( P > 0.05). Conclusion The Chinese materia medica for benefiting qi for activating blood circulation can reduce NT-proBNP and cTnI level, therefore alleviating the process of heart remodeling in order to improving the cure of chronic heart failure.
