七叶通脉胶囊对大鼠脑缺血再灌注模型的影响 被引量:6

摘要 目的观察七叶通脉胶囊对脑缺血再灌注动物模型体外血栓、血小板聚集功能、血液流变性的影响。方法选取大鼠240只,随机分为6组:空白组、脑缺血再灌注模型组、阳性对照血塞通胶囊42 mg/kg组、七叶通脉胶囊42、21、10.5 mg/kg剂量组。连续给药7 d后,制备脑缺血再灌注模型(空白组除外),缺血2 h灌注22 h后处死动物,采血,以全自动血流变快测仪测定血液流变性,体外血栓仪测定体外血栓,血液凝集仪测定血小板聚集。结果七叶通脉胶囊高、中、低剂量组给药后使脑缺血再灌注模型大鼠的血栓湿重、干重、血栓长度显著减小(P <0.01,P <0.05);血小板聚集率结果显示,高、中、低剂量组在1 min、3 min、5 min时和最大聚集率明显降低(P <0.01,P <0.05);血液流变性结果显示,七叶通脉胶囊高、低剂量组使高、中、低切的全血粘度均明显减小,中、低剂量组也使血浆粘度明显降低(P <0.05)。结论七叶通脉胶囊能通过改善缺血再灌注大鼠的血液流变性、抑制血栓形成和血小板聚集程度起到治疗脑缺血的作用。故推测活血化瘀作用可能也是七叶通脉胶囊治疗脑缺血的作用机制之一。 Objective To observe the effects of Qiye Tongmai Capsule on thrombus, platelet aggregation and hemorheology of rats with cerebral ischemia reperfusion. Methods The rats were randomly divided into 6 groups: normal group, cerebral ischemia reperfusion model group, Xuesaitong Capsule group with dose of 42 mg/kg, Qiye Tongmai Capsule groups with doses of 42 mg/kg, 21 mg/kg, 10.5 mg/kg. After 7 days of continuous administration, the model of cerebral ischemia reperfusion was prepared (except the normal group). After ischemia 2 h and reperfused 22 h, the animals were killed and blood was collected. The blood rheology,the thrombus in vitro and platelet aggregation were measured. Results After given three doses of Qiye Tongmai Capsule, the wet weight, dry weight and the length of thrombus of the rats in cerebral ischemia reperfusion model decreased significantly ( P <0.01, P <0.05). The results of platelet aggregation rate showed that the maximum aggregation rate and platelet aggregation rate of high, medium and low dose groups decreased significantly at 1 min, 3 min and 5 min ( P < 0.01, P < 0.05). The results of hemorheology showed that the whole blood viscosity of high, medium and low shear rates were significantly decreased in the high and low dose groups of Qiye Tongmai Capsule, and the plasma viscosity of middle and low dose groups was also significantly decreased ( P <0.05). Conclusion Qiye Tongmai Capsule can treat cerebral ischemia by improving the hemorheology, inhibiting thrombosis and platelet aggregation in rats with cerebral ischemia. Therefore, it is speculated that activating blood circulation and removing blood stasis may be one of the mechanisms of Qiye Tongmai Capsule in treating cerebral ischemia.
