摘要
以海南腰果为原料,通过碱溶酸沉法制备腰果蛋白,对其氨基酸的组成进行了分析,并通过酶解法制备腰果蛋白多肽,依次经过透析、柱层析分离纯化来研究腰果蛋白多肽的血管紧张素酶(Angiotensin Converting Enzyme,ACE)抑制活性。实验结果表明,腰果蛋白中富含必需氨基酸与非必需氨基酸,是一种优质蛋白资源,腰果蛋白中疏水性氨基酸含量为20.4g/100g,有助于其ACE抑制活性。利用碱性蛋白酶进行酶解后制得的酶解液(1mg/mL)ACE抑制率为71.24%±5.96%,经过透析后所得小分子组分[(0~3500)u]的ACE抑制率(1 mg/mL)为85.01%±1.44%,经葡聚糖凝胶(Sephadex G-25)分离纯化后,得到6种组分,其中ACE抑制活性最高的组分为P4,1 mg/mL P4的ACE抑制率为90.01%±0.16%[IC_(50)为(36.25±0.12)μg/m L],将P4进行反向高效液相色谱进一步分离纯化后,得到3个组分,其中P4-3的ACE抑制活性最高,1 mg/mL P4-3的ACE抑制率为91.04%±0.31%[IC_(50)为(2.12±0.067)μg/mL],表现出较高的ACE抑制活性,腰果蛋白ACE抑制肽有望应用于高血压的预防与功能食品的开发,对腰果中蛋白资源的开发利用有一定的实验意义。
In this paper, cashew protein were prepared from the cashews of Hainan province by alkali solubilization acid precipitation method and the amino acid composition was analyzed. The cashew protein polypeptide was prepared by enzymatic hydrolysis, followed by dialysis followed by separation and purification by column chromatography to study the angiotensin converting enzyme(ACE) inhibitory activity of the cashew protein polypeptide. The experimental results show that cashew protein is rich in essential amino acids and non-essential amino acids, and is a high-quality protein resource. The content of hydrophobic amino acids in cashew protein is 20.4 g/100 g, which contributes to its ACE inhibitory activity. The inhibition rate of ACE in the enzymatic hydrolysate(1 mg/mL) prepared by alkaline protease was 71.24%±5.96%, and the ACE inhibition rate of the small molecule fraction [(0~3500)u]obtained after dialysis was 85.01%±1.44% of 1 mg/mL. After separation and purification by Sephadex G-25, six components were obtained. The highest ACE inhibitory activity was P4. The inhibition rate was 90.01%±0.16% [IC50 was(36.25±0.12)μg/mL] of 1 mg/mL P4. After P4 was further separated and purified by reversed-phase high-performance liquid chromatography, three components were obtained, of which P4-3 had the highest ACE inhibitory activity, The ACE inhibitory rate of P4-3 in 1 mg/mL was 91.04%±0.31% [IC50 was(2.12±0.067)μg/mL], which showed a higher ACE inhibitory activity. Cashew protein ACE inhibitory peptide is expected to be used in the prevention of hypertension. The development of functional food has certain experimental significance for the development and utilization of protein resources in cashew nuts.
