不同途径给予胶原蛋白肽对免疫抑制小鼠免疫功能的影响 被引量:4

Effects of collagen peptides with different administration routes on immune functions of immunosuppressed mice

摘要 目的寻找胶原蛋白肽提高小鼠免疫功能的最佳给药途径。方法将小鼠随机分为灌胃组和腹腔注射组,两组均再分为对照组、模型组和100、300、600 mg胶原蛋白肽组,共计10个小组,每组6只小鼠。模型组和胶原蛋白肽组均采用糖皮质激素诱导小鼠免疫抑制,灌胃组和腹腔注射组的胶原蛋白肽组小鼠分别采用灌胃和腹腔注射的方式,连续7 d给予100、300、600 mg胶原蛋白肽/kg体重,对照组和模型组小鼠同时给予等渗盐水,于第8天处死小鼠,分别检测胸腺和脾脏指数、外周血白细胞分类及计数、CD4/CD8比值以及脾脏T淋巴细胞增殖能力。结果与模型组相比,腹腔注射胶原蛋白肽从低剂量组或中剂量开始即对小鼠脾脏指数、中性粒细胞百分比及绝对值、淋巴细胞百分比及CD4+/CD8+比值和脾脏淋巴细胞增殖能力均具有显著改善作用(P <0. 05),而且部分指标与对照组无显著差异(P> 0. 05)。而灌胃组只有600 mg胶原蛋白肽组对上述除脾脏指数之外的指标有显著改善作用(P <0. 05),但与对照组比较,差异仍有统计学意义(P <0. 05)。结论腹腔注射胶原蛋白肽对免疫抑制小鼠免疫功能的改善作用优于灌胃。 Objective To find out the best administration route of collagen peptides to improve the immune functions of mice. Methods Mice were randomly divided into 2 groups: intragastric administration group and intraperitoneal injection group. Then, the two groups were respectively divided into 5 subgroups: control group, model group and 100, 300, 600 mg collagen peptides groups, 6 mice in each group. Immunosuppression were induced in mice of model groups and collagen peptides groups with glucocorticoid. Mice in collagen peptides groups of intragastric administration group and intraperitoneal injection group were respectively intragastrically and intraperitoneally administrated 100, 300, 600 mg collagen peptides per kilogram by for 7 days. The mice were killed on the tenth days. Thymus and spleen indexes, white cells count and classification, the ratios of CD4 to CD8 and T lymphocyte proliferations of spleens were respectively analyzed. Results Compared with control group, the spleen indexes, numbers and percentages of leukocytes and percentages of lymphocytes and ratios of CD4+ to CD8+ were improved in mice after intraperitoneally administrated with different dose of collagen peptides from low or middle dose (P<0.05) and were not significantly different with control group (P>0.05). The above parameters except spleen indexes were improved in mice of 600 mg collagen peptides groups after intragastric administration (P<0.05), but they were still significantly different with control group (P<0.05). Conclusion Intraperitoneal injection is superior to intragastric administration with collagen peptides in improvements of the immune functions in immunosuppressed mice.
