环己酮二腙致C57BL/6J小鼠脑白质脱髓鞘病变及髓鞘再生的雌雄差异性研究 被引量:2

摘要 目的探讨环己酮二腙(CPZ)诱导C57BL/6J小鼠脑白质脱髓鞘病变及自发性髓鞘再生的雌雄差异。方法 48只C57BL/6J小鼠,雌雄各半,随机选取32只为造模组,16只为对照组(A组),对照组常规饲养,造模组用含CPZ饲料饲养6周,造模成功后再随机分为脱髓鞘组(B组)和髓鞘再生组(C组)。A组和B组于6周末行运动协调能力测试后处死取脑组织,C组继续常规饲养6周后行运动协调能力测试后处死取脑组织,所有脑组织均行卢卡斯快蓝(LFB)染色,检测髓鞘碱性蛋白(MBP)和脑细胞凋亡比值。结果与A组比较,B组运动协调能力及MBP含量明显降低(P <0. 05),脑细胞凋亡比值及脱髓鞘病变程度评分明显增加(P <0. 05)。C组运动协调能力及MBP含量较B组显著提高(P <0. 05),脑细胞凋亡比值及脱髓鞘病变程度评分较B组低(P <0. 05)。B组组内雌雄比较运动协调能力、脱髓鞘病变程度评分、脑细胞凋亡比值、MBP含量差异均无统计学意义(P> 0. 05)。C组组内比较,雌组比雄组运动协调能力好、脱髓鞘病变程度评分低及MBP含量高,脑细胞凋亡比值差异无统计学意义(P> 0. 05)。结论 CPZ致C57BL/6J小鼠脑白质脱髓鞘病变无雌雄差异,在髓鞘再生期,雌性小鼠具有更好的髓鞘再生能力。
