摘要
目的:研究牛蒡子中总木脂素的提取、纯化工艺、确定牛蒡子中总木脂素纯化前后体外抑菌作用。方法:以牛蒡苷为指标,采用单因素和正交实验确定牛蒡子总木脂素的最佳提取工艺,利用大孔吸附树脂法对牛蒡子有效成分进行纯化工艺研究,采用牛津杯法对纯化前后的牛蒡子有效成分进行抑菌实验研究。结果:牛蒡子最佳提取工艺为用80%乙醇为提取溶剂,提取3次,加醇量为12倍,每次1.5h。最佳纯化工艺为AB-8型大孔吸附树脂装柱,0.05g·mL^(-1)上样浓度,6BV的蒸馏水和5BV的70%乙醇洗脱,70%乙醇洗脱液洗脱部分为牛蒡子总木脂素。牛蒡子总木脂素对金黄色葡萄球菌和肺炎链球菌有抑菌作用。结论:牛蒡子总木脂素经本工艺提取、纯化后,其纯度可达65.27%以上,简单易行,适合工业化生产,牛蒡子总木脂素对于金黄色葡萄球菌和肺炎链球菌具有抑菌作用,为进一步研究其有效成分治疗急慢性咽炎等咽喉疾病提供前期试验基础。
Objective:To study the extraction and purification process of total lignans from Arctium lappa L.and to determine the antibacterial activity of total lignans in burdock.Methods:Using Burdock as an indicator,the best extraction process of total Lignan from Burdock was determined by single factor and orthogonal experiments,Purification of the active constituents of Arctium lappa L.by macroporous adsorption resin,The bacteriostasis experiment was carried out on the active constituents of the burdock before and after purification by the Oxford Cup method. Results:The best extraction process of burdock is to extract 80 times with 80%ethanol as extraction solvent,and add 12 times of alcohol,1.5 hours each time.The best purification process is AB-8 type macroporous adsorption resin loading column,0.05g·mL^-1 loading concentration,6BV distilled water and 5BV 70% ethanol eluting,70% ethanol eluent eluting part of burdock total wood Lipoprotein.The total lignans of Burdock have antibacterial activity against Staphylococcus aureus and Streptococcus pneumoniae.Conclusion:After the extraction and purification of the total lignans of Burdock,the purity of the lignans can reach 65.27% or more,which is simple and easy to be industrialized,Burdock total lignans have antibacterial activity against Staphylococcus aureus and Streptococcus pneumoniae,To provide further preliminary research basis for further study of its active ingredients in the treatment of throat diseases such as acute and chronic pharyngitis.
