摘要
目的分析2014年以来辽宁省流行的水痘-带状疱疹病毒(varicella-zoster virus,VZV)基因型别,区分疫苗株与野毒株,为水痘防控奠定理论基础。方法应用PCR法对VZV基因组第22个开放阅读框(open reading frame22,ORF22)基因进行特异性扩增,并对PCR产物进行序列测定,结合单核苷酸多态性(single nucleotide polymorphism,SNP)方法确定病毒的基因型别。扩增ORF38、ORF54、ORF62基因,测序后寻找目标片段中是否存在相应限制性酶切位点,以区分疫苗株与野毒株。结果辽宁省2014~2018年共采集到68份临床诊断水痘病例的疱疹液样本,其中VZV-013-2015样本序列与DUMAS株一致,为Clade 1基因型,其余样本序列均为Clade 2基因型,与疫苗株Voka一致。辽宁省VZV分离株在69349位点PstⅠ(+),95241位点BglⅠ(+),106262位点SmaⅠ(-)。结论 Clade 2基因型为辽宁省VZV病毒流行优势基因型;辽宁省VZV均为野毒株感染。
Objective To understand the genotypes of varicella zoster virus(VZV)prevalent in Liaoning Province,China since 2014,distinguish vaccine strains from wild strains,and provide a basis for prevention and control of varicella.Methods The fragment of open reading frame(ORF)22 gene was amplified by PCR and sequenced,based on which the genotype of virus was determined by analysis of single nucleotide polymorphism(SNP). ORF38,ORF54 and ORF62 genes were amplified and sequenced,in which the corresponding restriction sites were searched to distinguish the vaccine strains and wild strains. Results A total of 68 clinic specimens of VZV were collected during 2014 ~ 2018. Except for VZV-013-2015 was of genotype Clade 1,the other specimens were of genotype Clade 2,which were consistent with those of vaccine strain Voka. The sites 69349 and 95241 of VZV isolates in Liaoning Province showed were restriction sites of PstⅠand BglⅠrespectively,while site 106262 was not the restriction site of SmaⅠ. Conclusion Clade 2 genotype was the dominant genotype of VZV in Liaoning Province. All the VZV isolates in Liaoning Province were wild strains.
