摘要
采用一步法合成了氨基化MNPs,以其为磁源,制备了海藻糖合酶M-CLEAs,考察了M-CLEAs的制备条件和酶学性质.结果表明,当酶蛋白与壳聚糖修饰的磁性粒子(MNPs-CS1)、戊二醛及壳聚糖浓度比分别为1∶4、1∶1、1∶2时,所得海藻糖合酶M-CLEAs的酶活回收率最高,达到68%.海藻糖合酶M-CLEAs的最适反应温度为60℃,最适反应p H为7.0,对热和p H的耐受性均优于游离海藻糖合酶,并且具有较好的重复使用性能.
The chitosan-modified magnetic particles (MNPs-CS1) were synthesized using one-step synthesis and the trehalose synthase magnetic cross-linked enzyme aggregates (M-CLEAs) were prepared using MNPs-CS1. The preparation conditions and catalytic properties of trehalose synthase M-CLEAs were investigated. The results showed that the optimized preparation condition was the ratio of enzyme protein to chitosan-modified magnetic particles (MNPs-CS1), glutaraldehyde, chitosan were 1∶4, 1∶1, 1∶2. Under this condition, the activity recovery of trehalose synthase M-CLEAs was reached up to 68%. The optimum catalytic temperature and pH of trehalose synthase M-CLEAs were 60 ℃ and 7.0. The temperature and pH resistance and reusability of trehalose synthase M-CLEAs were better than that of the free enzyme.
