miR-93对神经母细胞瘤细胞侵袭和迁移的影响及机制研究 被引量:1

摘要 目的研究miR-93对神经母细胞瘤细胞侵袭和迁移的影响及机制。方法将2017年1月至2017年12月在山东省德州市人民医院接受治疗的小儿神经母细胞瘤患者13例作为研究对象,取患者肿瘤原发部位以及转移部位的组织样本,利用PCR扩增的方法,检测所取样本中miR-93的表达水平。并在神经母细胞瘤细胞SH-N-SH和SH-SY5Y细胞中对miR-93进行转染,在转染后的24 h利用PCR扩增技术检测其中miR-93的表达水平。并利用Transwell侵袭实验对过表达miR-RAN后细胞侵袭能力的影响。结果转移部位miR-93表达水平显著低于原发部位,差异有统计学意义(P<0.05)。转染24 h后,利用实施定量PCR检测SH-N-SH和SH-SY5Y中miR-93的表达水平,结果转染后的miR-93水平要显著高于对照组,差异有统计学意义(P<0.05)。对Transwell细胞侵袭实验能力进行比较,与对照组相比,进行转染后的miR-93过表达质粒组的细胞穿膜数量显著降低,差异有统计学意义(P<0.05)。说明过表达的miR-93能够显著抑制SH-N-SH和SH-SY5Y两种细胞的侵袭能力。结论miR-93能够抑制神经母细胞瘤的侵袭和转移,从而作为神经母细胞瘤的治疗靶标。 ObjectiveTo investigate the effect of miR-93 on invasion and migration of neuroblastoma cells and its mechanism.MethodsFrom January 2017 to December 2017, 13 children with neuroblastoma treated in the People's Hospital of Dezhou were selected as the objects. The original site of the tumor and the tissue samples of the metastatic site were taken. The expression of miR-93 in the samples was detected by PCR amplification. MiR-93 was transfected in SH-N-SH and SH-SY5Y cells of neuroblastoma cells, and the expression level of miR-93 was detected by PCR amplification after transfection. The effect of Rranswell invasion on cell invasiveness after overexpression of miR-RAN was also investigated.ResultsThe expression level of miR-93 in the metastatic site was significantly lower than that in the original site, and the difference was significant(P<0.05). After transfection of 24 h, the expression level of miR-93 in SH-N-SH and SH-SY5Y was detected by quantitative PCR. The results showed that the level of miR-93 after transfection was significantly higher than that of the control group, and the difference was significant(P<0.05). Compared with the control group, the number of Transwell cells in the miR-93 overexpressed plasmid group was significantly lower than that in the control group, the difference was significant (P<0.05). The overexpression of miR-93 could significantly inhibit the invasion ability of SH-N-SH and SH-SY5Y two kinds of cells.ConclusionsMiR-93 can inhibit the invasion and metastasis of neuroblastoma, which can be used as a target for the treatment of neuroblastoma.
