摘要
目的研究薏苡的茎和叶的石油醚萃取部位分离组分的抗肿瘤活性。方法薏苡茎叶的乙醇提取物经石油醚萃取后,以不同比例的石油醚-乙酸乙酯为流动相将石油醚相用硅胶柱色谱分离,所得洗脱液以乙酯比例10∶1石油醚-乙酸为展开剂用薄层色谱分析,合并相同极性段,各极性段经甲醇结晶处理共得到4个分离组分(样品1,2,3,4号),用CCK-8法检测这4个分离组分对3种肿瘤细胞株体外生长的抑制作用,并计算其半数抑制浓度(IC_(50)值)。结果从石油醚-乙酸乙酯比例20∶1的洗脱剂中得到了样品1号和4号,从石油醚-乙酸乙酯比例15∶1的洗脱剂中得到了样品2号和样品3号,4个样品均为干燥的混合物。4个样品对3种肿瘤细胞株的体外生长均有抑制作用,其中3号分离组分的抗肿瘤活性最好,3号样品对人宫颈癌(He La)、肝癌(Hep G2)、胃癌(SGC-7901)等细胞株的IC_(50)值分别为126. 1,145. 5,129. 2μg·m L^(-1)。结论薏苡的茎、叶乙醇提取物的石油醚萃取部位分离组分具有一定的抗肿瘤作用,其中3号样品抗肿瘤活性最好。
Objective To explore the antitumor effects of isolated components of petroleum ether extracts from stalks and leaves of Coix.Methods The ethanol extracts from the stems and leaves of Coix were extracted by petroleum ether.The petroleum ether extracts were separated by silica gel column chromatography with different proportions of petroleum ether-ethyl acetate as mobile phase.The eluents were analyzed by thin layer chromatography(TLC)with petroleum ether-ethyl acetate ratio of 10∶ 1 as unfolding agent.The same polar segments were merged and four fractions(sample 1,2,3,4)were obtained by methanol crystallization.The in vitro growth inhibitory effects of the four fractions(sample 1,2,3,4)on 3 kinds of tumor cell lines was detected by Cell Counting Kit-8(CCK-8)assay.The half inhibitory concentration(IC50 value)was calculated.Results Sample 1 and sample 4 were obtained from the eluent with petroleum ether-ethyl acetate ratio of 20:1.Sample 2 and sample 3 were obtained from the eluent with petroleum ether-ethyl acetate ratio of 15:1.All the four samples were dry mixtures.The four samples can inhibit the growth of 3 kinds of tumor cell lines in vitro,and the No.3 isolate has the best anti-tumor activity.The IC50values of No.3 isolate on human cervical cancer(HeLa),liver cancer(HepG2)and gastric cancer(SGC-7901)cell lines were 126.1,145.5 and 129.2 μg·mL-1,respectively.Conclusion The isolated components of petroleum ether extracts from stalks and leaves of Coix have certain antitumor effect,the antitumor activity of No.3 isolate was the best.
