摘要
目的:探讨丹参通脉方对同型半胱氨酸(Hcy)诱导的血管平滑肌细胞(VSMC)增殖、迁移的影响及其机制。方法:取SD大鼠胸主动脉VSMC传代培养后分为对照组、Hcy组、丹参通脉方低剂量组、高剂量组和瑞舒伐他汀组。采用MTT检测细胞凋亡水平,采用Transwell法检测VSMC迁移能力,采用Western-blotting检测NF-κB、CHOP和TRB3蛋白表达水平。结果:与对照组比较,Hcy组细胞OD值、细胞迁移面积和穿透细胞数量显著上升,差异有统计学意义;与Hcy组比较丹参通脉方低剂量组、高剂量组和瑞舒伐他汀组OD值、细胞迁移面积和穿透细胞数量显著下降,且丹参通脉方高剂量组显著低于丹参通脉方低剂量组。与对照组比较Hcy组NF-κB、CHOP和TRB3表达水平显著上升,与Hcy组比较丹参通脉方低剂量组、高剂量组和瑞舒伐他汀组NF-κB、CHOP和TRB3表达水平显著下降,且丹参通脉方高剂量组显著低于丹参通脉方低剂量组。结论:丹参通脉方可能是通过ERS/NF-κB发挥抑制Hcy诱导VSMC增殖和迁移的作用。
Objective: To investigate the effect of Danshen Tongmai Recipe on Proliferation and migration of vascular smooth muscle cells(VSMCs) induced by homocysteine(Hcy) and its mechanism. Methods: SD rat thoracic aorta VSMC after subculture. The cells were divided into control group, Hcy group(500 μmol/L Hcy), low dose group(500 μmol/L Hcy + 1 mg/L salvia miltiorrhiza), high dose group(500 μmol/L Hcy + 10 mg/L Salvia miltiorrhiza) and rosuvastatin group(500 μmol/L Hcy + 1 mg/L rosuvastatin). MTT assay was used to detect the apoptosis level. The migration ability of VSMC was detected by Transwell method. The expression of NF-κB, CHOP and TRB3 Protein was detected by Western-blotting. Results: Compared with the control group, the 24, 48 h OD value, cell migration area and the number of penetrating cells in the Hcy group were significantly increased. Compared with the Hcy group, 24, 48 h OD value, cell migration area and the number of Penetrating cells in the low dose group, the high dose group and rosuvastatin group were significantly lower, and the same between low dose group and the high dose group. Compared with the control group, the expression of NF-κB, CHOP and TRB3 in Hcy group was significantly higher than that in control group. Compared with Hcy group, the expression of NF-κB, CHOP and TRB3 in the low dose group, the high dose group and rosuvastatin group were significantly lower, and the same between low dose group and the high dose group. Conclusion: Danshen Tongmai Recipe may inhibit Hcy-induced VSMC proliferation and migration through ERS/NF-κB.
