摘要
目的观测氨基葡萄糖硫酸盐、白芍提取物与硫酸软骨素联合制备的混合物对大鼠骨密度的作用。方法雌性Wistar大鼠50只,按体重随机分为5组,每组10只,其中40只雌性大鼠以切除双侧卵巢的方法建立骨密度低下模型,分别为去卵巢低0.16 g/(kg·bw)、中0.32 g/(kg·bw)、高0.96 g/(kg·bw)剂量组和去卵巢对照组,另设假手术对照组。实验组分别给予不同剂量的氨基葡萄糖硫酸盐、白芍提取物与硫酸软骨素混合物,去卵巢对照组和假手术对照组均经口给予相应容积去离子水,每日1次灌胃,连续3个月后检测大鼠股骨重量、骨密度、骨钙含量以及血钙水平。观察动物生长情况以及对体重的影响。结果去卵巢对照组大鼠的股骨钙含量及骨密度均显著低于假手术对照组(P<0.01),成功建立大鼠骨密度低下模型。各剂量组的大鼠的股骨钙含量、股骨中点骨密度和远心端骨密度均显著高于去卵巢对照组(P<0.05,P<0.01)。各剂量组血钙水平与去卵巣对照组比较无统计学差异(P>0.05)。结论氨基葡萄糖硫酸盐、白芍提取物与硫酸软骨素混合物具有增加大鼠骨密度的功能。
Objective To study the effects of mixture of glucosamine sulfate,white peony root extract and chondroitin sulfate on rats’ bone mineral density.Methods According to weight,a total of 50 healthy female Wistar rats were randomly assigned to five groups.Among them,the ovariectomized(OVA) rats control group and low-,medium-,high-dose mixture-treated groups [0.16,0.32,0.96 g/(kg·bw) ]were established the bone mineral density reducing model by resecting with bilateral ovaries.The another group was the sham operation control group.The three experimental groups were given different dose of mixture and the sham operation control group,OVA control group were administrated by gavage deionized water once a day for 3 months.At the end of the experiment,taking the femur after anesthesia,according to the follow-up experiments need treatment.The weight,bone mineral density of the femur,bone calcium content and serum calcium were measured and the impact of rats’ growth on weights was observed.Results The bone calcium content and bone density of the OVA control group were significantly lower than that of sham operation group(P<0.01),and the bone mineral density reducing model was set up successfully.The bone calcium content,the midpoint and distal end of femur density in each dose groups were significantly higher than that of the OVA control group(P<0.05,P<0.01).Serum calcium levels of each dose group had no statistical difference compared with OVA control(P>0.05).Conclusion The mixture of glucosamine sulfate,white peony root extract and chondroitin sulfate has the function of increasing bone mineral density.
