肝癌组织LncRNA TINCR和ANRIL表达水平及其与患者预后的关系 被引量:11

Expressions of LncRNA TINCR and ANRIL in liver cancer tissues and their relationships with the prognosis of the patients

摘要 目的探讨肝癌组织长链非编码RNA(LncRNA)组织分化诱导非蛋白质编码RNA(TINCR)和细胞周期激酶抑制因子4基因座中反义非编码RNA(ANRIL)表达水平及其与患者预后的关系。方法选取2010年7月至2012年11月本院进行手术切除的68例肝癌患者的肿瘤组织及癌旁组织,采用RT-PCR方法检测肝癌组织和癌旁组组中LncRNA TINCR和ANRIL相对表达量,分析LncRNA TINCR和ANRIL表达与患者临床病理特征及生存率的关系。结果肝癌组织中TINCR和ANRIL相对表达量均显著高于癌旁组织(P<0.05);TINCR表达量与患者肿瘤大小、肿瘤分化程度及TNM分期有关,ANRIL表达量与患者肿瘤大小和TNM分期有关;TINCR高表达患者无瘤生存率及5年总生存率均显著低于TINCR低表达患者(P<0.05);ANRIL高表达患者无瘤生存率及5年总生存率均显著低于ANRIL低表达患者(P<0.05);肿瘤大小、TINCR高表达和ANRIL高表达是影响肝癌患者预后的危险因素。结论肝癌组织中TINCR和ANRIL表达显著上调,可能参与肿瘤进展过程,与患者预后密切相关。 Objective To study the expressions of long non-coding RNA( LncRNA), tissue differentiation inducing ncRNA( TINCR) and antisense non-coding RNA in the INK4 locus(ANRIL) in liver cancer tissues and their relationships with the prognosis of the patients. Methods From July 2010 to November 2012, tumor tissues and para cancerous tissues of 68 patients with liver cancer were surgically removed and collected. RT-PCR method was used to detect the relative expression levels of LncRNA, TINCR and ANRIL in tumor tissues and para cancerous tissues;the relationships between the expressions of LncRNA, TINCR and ANRIL with the clinicopathological features and survival rate of patients were analyzed.Results The relative expressions of TINCR and ANRIL in HCC tissues were significantly higher than those in para cancerous tissues(P<0.05);the expression of TINCR was related to tumor size, tumor differentiation and TNM stage;the expression of ANRIL was related to tumor size and TNM stage;the tumor free survival rate and the 5-year overall survival rate of patients with high TINCR expression were significantly lower than those of patients with low TINCR expression( P<0.05);the tumor free survival rate and the 5-year overall survival rate of patients with high ANRIL expression were significantly lower than those of patients with low ANRIL expression(P<0.05);tumor size, high expression of TINCR and high expression of ANRIL were risk factors of prognosis in patients with liver cancer. Conclusion The expressions of TINCR and ANRIL were significantly up-regulated in HCC, which might be involved in the process of tumor progreddion,and was closely related to the prognosis of patients.
