摘要
目的建立高效液相荧光检测(HPLC-FLD)法测定复方五指毛桃颗粒中补骨脂素的含量。方法采用YMC-Pack-AM色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-水(34∶66),等度洗脱;流速为1.0 m L·min^(-1);FLD检测器的激发波长(Ex)为247 nm,发射波长(Em)为441 nm;柱温为30℃。结果补骨脂素进样量在3.117 0~2 992.200 0 ng(r=0.999 8)范围内与峰面积呈良好的线性关系,平均加样回收率为99.77%(n=9),RSD为1.38%。结论本研究所建立的HPLC-FLD法简便、准确、稳定,可用于复方五指毛桃颗粒中补骨脂素的含量测定,为复方五指毛桃颗粒的质量评价提供了参考。
Objective To develop a high performance liquid chromatography fluorescence detection(HPLC-FLD)method for determination of psoralen in compound Fici hirtae radix granule.Methods Chromatographic separation was carried out on a YMC-Pack AM column(250 mm×4.6 mm,5 μm) with a mobile phase consisting of acetonitrile and water(34∶66)by isocratic elution at a flow rate of 1 m L·min^-1.The maximum excitation wavelength and emission wavelength of the fluorescence detector was set at 247 nm and 441 nm,and the column temperature was maintained at30 ℃.Results The calibration curves of psoralen showed good linear relationship within the range of 3.117 0-2992.200 0 ng(r=0.999 8),and the average recovery of the psoralen was 99.77%(n=9),the relative standard deviation(RSD) was 1.38%.Conclusion The established HPLC-FLD method is simple,rapid,accurate and stable for the determination of psoralen in compound Fici hirtae radix granule,which will supply evidence for the development and quality control of compound Fici hirtae radix granule.
