摘要
目的探讨HOTAIR对宫颈癌HeLa细胞增殖及自噬的影响。方法应用qRT-PCR法检测宫颈癌和正常宫颈组织中HOTAIR表达水平,并分析其与临床病理参数的关系。构建宫颈癌He La细胞HOTAIR过表达细胞株(pcDNA3.1(+)-HOTAIR组)和HOTAIR干扰细胞株(si-HOTAIR组),CCK-8法检测HOTAIR对HeLa细胞增殖的影响。Western blot和免疫荧光分别检测各组细胞自噬关键基因LC3的蛋白和荧光表达。结果宫颈癌组织HOTAIR表达水平显著高于对照组(P <0.001),而与临床病理参数的关系无统计学意义(P>0.05)。与对照组相比,pcDNA3.1(+)-HOTAIR组HeLa细胞增殖水平显著升高(P <0.001),细胞质内LC3点状荧光明显增加。结论 HOTAIR在宫颈癌组织中表达增高,同时可上调宫颈癌HeLa细胞的自噬水平并促进HeLa细胞增殖,其有望成为判断宫颈癌恶性程度和患者预后的生物学标志物。
Objective To investigate the effect of HOTAIR on proliferation and autophagy in cervical cancer He La cells. Methods q RT-PCR were used to measure the expressions of HOTAIR in cervical cancer and normal cervix tissues, and analyzed the correlation between HOTAIR expressions and clinicopathological parameters. The over-expression of HOTAIR HeLa cells called pcDNA3.1(+)-HOTAIR group and the inhibition of HOTAIR HeLa cells called si-HOTAIR group. The proliferation of HeLa cells were measured by CCK-8 assay. The expressions of autophagy related gene LC3 were assessed by western blot and immunoìuorescence assay respectively. Results The HOTAIR expression was higher in cervical cancer tissues than in normal tissues(P < 0.001), but without significant relationship with clinical parameters. Compared with control groups, the proliferation was promoted in pcDNA3.1(+)-HOTAIR group(P < 0.001). The LC3 puncta throughout the cytoplasm were increased as well. Conclusions Lnc RNA HOTAIR was highly expressed in cervical cancer and could upregulate the autophagy and promote the proliferation of cervical cancer HeLa cells. It could be a potential biomarker for estimating the malignancy and prognosis of cervical cancer.
