摘要
目的建立HPLC法同时测定山茱萸药材中莫诺苷、当药苷、马钱苷和山茱萸新苷I含量的方法,并比较不同部位中4种成分的差异。方法采用Diamonsil Plus C_(18)-A色谱柱(250 mm×4.6 mm,5μm),流动相乙腈-0.3%磷酸溶液梯度洗脱,检测波长240 nm和270 nm。结果 4种成分在山茱萸果肉、果梗和果核中的含量存在显著差别。果肉中莫诺苷和马钱苷的含量显著高于果梗及果核,当药苷在果核中含量最高,山茱萸新苷I在果梗中含量最高。结论山茱萸不同部位化学成分存在显著差异,建议去核入药。
Objective To establish an HPLC method for simultaneous determination of morroniside, sweroside, loganin and cornuside Ⅰin Corni Fructus and compare the differences of the four components among different parts. Methods The separation is performed on a Diamonsil Plus C18-A column(250 mm × 4.6 mm, 5 μm) with a gradient elution of acetonitrile and 0.3% phosphoric acid solution, and the detection wavelengths are set at 240 nm(morroniside, sweroside, loganin) and 270 nm(cornuside I). Results There are significant differences in the contents of four components among pulp, carpopodium and fruit stone of Corni Fructus. The contents of morroniside and loganin in the pulp are obviously higher than in carpopodium and fruit stone. The content of sweroside is the highest in the fruit stone, and the content of cornuside I is the highest in the carpopodium. Conclusion There are significant differences in the contents of four components among different parts of Corni Fructus, it is suggested that the fruit stone should be removed when Corni Fructus is used as medicine.
