摘要
目的 :研究糖皮质激素对人类糖皮质激素受体α和 βmRNA表达的调节作用。 方法 :采用定量逆转录 -聚合酶链反应 (RT -PCR)检测受体mRNA。结果 :人骨肉瘤细胞系、卵巢癌细胞系及外周血白细胞中除有糖皮质激素受体αmRNA表达外 ,还有糖皮质激素受体 βmRNA的表达 ;糖皮质激素对糖皮质受体α和 βmRNA的表达均具有明显的降调作用 ,且具有剂量依赖性和时间依赖性特点。结论 :糖皮质激素受体 β在人类多种组织细胞中均有表达 。
AIM: To clarify the effects of glucocorticoids on the messenger RNA expression of glucocorticoid receptor (GRα and GRβ). METHODS: messenger RNA of GR α and GRβ were determined by quantitive RT-PCR. RESULTS: Besides GRα mRNA, GRβ mRNA is also expressed in human osteosarcoma cell line HOS-8603, human ovarian carcinoma cell line 3AO and human peripheral white blood cells. After administration of dexamethasone, both GRα mRNA and GRβ mRNA were down-regulated in a time-dependent and dose-dependent manner. CONCLUSION: GRβ expresses in various kinds of human tissue cells. The expression of GRβ is regulated by glucocorticoids.
