摘要
目的 研究抗抑郁处理对慢性应激抑郁模型大鼠海马鸟苷酸结合蛋白 (G蛋白 )表达的影响。方法 将雄性Sprague Dawley大鼠 32只随机分为 5组 ,其中应激模型组 7只 ,阿米替林治疗组(阿米替林组 ) 7只 ,电针百会印堂穴组 (电针治疗组 ) 6只 ,电针对照点组 (电针对照组 ) 6只 ,正常对照组 (不施加应激处理 ) 6只。然后用Westernblotting方法检测各组大鼠海马 3种G蛋白亚型的含量。结果 慢性应激抑郁模型大鼠海马的Gαi、Gαq蛋白表达无明显变化 ;Gαo蛋白含量 (吸光度 )为 1 19±0 15 ,高于正常大鼠的 0 91± 0 14,P <0 0 5 ;阿米替林可使其降低至正常水平 ,而电针各组未见有明显降低作用。结论 阿米替林可能通过调节Gαo表达来发挥抗抑郁作用 ,电针百会印堂穴的抗抑郁机制可能与阿米替林不同 ,与 3种G蛋白亚型的表达无关。
Objective To explore the change of G protein content in chronic stress depression animal model after different antidepressive treatment. Methods Thirty-two male Sprague-Dawley rats were randomly divided into 5 groups, with 7 in stress model group, 7 in amitriptyline group (treated with amitriptyline), 6 in electrical acupuncture (YINTANG and BAIHUI points) group, 6 in non-point electro acupuncture group and 6 in normal control group. The G protein level in hippocampus was detected with Western blotting. Results Compared with normal rats, there was an increase of Gαo content in depression model rats with the relative value of Gαo (1.19±0.15) vs. (0.91±0.14) (P<0.05). Amitriptyline could decrease Gαo content to normal level, but electro acupuncture did not. Conclusion The effect of amitriptyline on Gαo may be its underlying antidepressive mechanism. The antidepressive mechanism of electro acupuncture may be different from amitriptyline in the central neurotransmission.
