清心滋肾汤对围绝经期大鼠卵巢Caspase-3 Caspase-9及Apaf-1的影响 被引量:5

Effects of Qingxin Zishen Decoction on the Expression of Caspase-3,Caspase-9 and Apaf-1 in Rat Ovaries During Perimenopausal Period

摘要 目的:研究中药清心滋肾汤对围绝经期大鼠卵巢Caspase-3、Caspase-9、Apaf-1的影响,探讨清心滋肾汤治疗绝经综合征的作用机制。方法:选用自然衰老的围绝经期雌性SD大鼠40只,月龄12月,随机分为4组(n=10):围绝经期模型组、清心滋肾汤低剂量组、清心滋肾汤中剂量组、清心滋肾汤高剂量组。另选4月龄的雌性SD大鼠10只作为青年对照组。连续灌胃4周后,采用Western blot(蛋白免疫印迹杂交)方法检测各组大鼠卵巢Caspase-3、Caspase-9、Apaf-1蛋白的表达,采用RT-PCR(逆转录-聚合酶链反应)检测大鼠卵巢Caspase-3、Caspase-9、Apaf-1 mRNA表达。结果:模型组大鼠卵巢Caspase-3、Caspase-9、Apaf-1蛋白及mRNA表达普遍高于青年组,清心滋肾汤各剂量组Caspase-3、Caspase-9、Apaf-1蛋白及mRNA表达呈不同程度的下降。结论:清心滋肾汤能下调卵巢Caspase-3、Caspase-9、Apaf-1凋亡因子的表达水平,延缓卵巢衰老,治疗绝经综合征。 Objective: To investigate the effects of Qingxin Zishen Decoction (QXZSD) on the expression of Caspase-3, Caspase-9 and Apaf-1 in rat ovaries during perimenopausal period. Methods: Fifty female 12 months old SD rats during perim- enopausal period were selected by unforced aging. Then the rats were divided into 5 groups randomly: the aged control group, the QXZSD low-dose, mid-dose, high-dose groups, the Remifemin group. Ten 4 months old female rats were selected as young controlled group. Intragastric administrations were conducted for 4 weeks continuously. The expression of Caspase-3, Caspase-9, Apaf-1 protein and mRNA in rat ovaries were detected by Western blot and RT-PCR, respectively. Results: The level of Caspase-3, Caspase-9, Apaf-1 protein and mRNA in rat ovaries was up regulated in the aged controlled group com- pared to the young controlled group; the expression of Caspase-3, Caspase-9, Apaf-1 protein and mRNA in QXZSD group was down regulated in different degrees, and it showed no difference between QXZSD high-dose and Remifemin group. Conclusion: QXZSD can down-regulate the expression of Caspase-3, Caspase-9, Apaf-1 protein and mRNA in rat ovaries of during perim- enopausal period, postpone the ovarian failure and cure perimenopausal syndrome.
