摘要
目的:研究三氧化二砷(As2O3)对鼠移植性肝癌端粒酶活性的影响。方法:建立小鼠HepA肝癌移植瘤模型(实体瘤模型),采用尾静脉注射As2O3,同时设生理盐水(NS)和5-氟尿嘧啶(5-FU)对照组,对比观察As2O3对肝癌实体瘤模型的生长抑制作用。利用端粒重复扩增实验(TRAP)结合非变性聚丙烯酰胺凝胶电泳银染法测定鼠移植性肝癌细胞端粒酶活性的变化。 结果:5—FU组和As2O3组肝癌荷瘤鼠皮下肿瘤的生长均受到明显抑制。NS组鼠肝癌细胞端粒酶活性不受影响,仍为强阳性;As2O32.0mg/kg组及5-FU组均未见明显抑制;As2O33.5mg/kg组端粒酶活性呈完全抑制。结论:As2O3对鼠移植性肝癌细胞端粒酶活性有抑制作用,有可能成为应用于临床的端粒酶抑制剂。
Objective: To study the influence of As2O3 on telomerase activity in the HepA solid liver tumor mice model. Methods: The HepA solid liver tumor mice model was built up, As2O3 was injected through mice tail vein. The negative con-trol group was injected normal saline(NS) and the positive control group was injected 5 - Fu. Combining PAGE - silver stain with TRAP, we detected the telomerase activity in HepA solid liver tumor mice when various concentration was given. Re-sults : After the administration of As2O3 and 5 - FU, the growth of solid tumor was obviously inhibited. The telomerase activity of NS control group was highly expressed while the telomerase activity of 3. 5mg/kg As2O3 group was totally inhibited, 2. 0mg/kg As2O3 group and 5 - FU group were not inhibited. Conclusion: As2O3 can greatly inhibit telomerase activity in the HepA solid liver tumor mice model,which will be one of the useful clinical anti-telomerase agents.
