摘要
目的观察类缺血后神经元内钙离子浓度及细胞活性的变化,探讨缺血性神经元的损伤与细胞内钙离子的关系及不同钙阻滞剂的作用。方法采用孕小鼠皮层神经元培养技术,应用相差显微镜观察培养的神经元形态结构,激光共聚焦扫描显微镜检测细胞内钙离子浓度。结果给予神经元类缺血处理10min后,神经元内钙离子浓度明显高于尼卡地平组(P<0.01)。类缺血处理10min后再灌注2h细胞损伤程度较尼卡地平干预组有显著性差异。结论尼卡地平可抑制神经元细胞内钙离子浓度的升高,降低缺血性神经元的损伤的程度。
Objective To study the changes of intracellular Ca2+ and cell activity in cortical neurons after ischemic like condition, and protection of nicardipine on the neurons. Methods Neurons were cultured and loaded with Flu 3/AM ,i was measured by fluorescent intensity (FI) in each cell with confocal microscopy, and the intensity of injured neurons was analyzed with MTT reduction assay.Result After 10 min ischemic like condition(deprived oxygen/glucose), a rapid increase in fluorescence intensity was observed, There was obvious difference between ischemic group and therapy group(P< 0.01). The OD value of injured neurons in ischemic group was significant lower than therapy group. Conclusion Nicardipine reduced the increase of i in cortical neurons and the intensity of the injured neurons after ischemic like condition.
