摘要
采用正交设计,探讨了影响甜柿试管苗生根的复合因素,结果表明:在添加细胞分裂素CPPU 0.2mg/L的MS(1/2N)继代培养基上,经适当时间继代培养的嫩梢经IBA 250 mg/L浸基部15 s后转接到1/2MS(1/2N)+活性炭1 000mg/L的生根培养基中或直接接种到添加有IBA 1 mg/L的上述生根培养基中诱导生根的效果好。不仅生根率高,根生长快,且根条数多,易于移栽成活。
The factors influencing rooting of nonastringent persimmon(Diospyros kaki L. f. ) shoots in vitro were studied by orthogonal design. The result showed that the shoots subcultured constantly on the MS(1/2N) medium supplemented with CPPU 0. 2 mg/L were dipped in IBA solution 250 mg/L for 15 s, and were incubated on the half-strengthen MS(1/2N) medium containing active carbon 1 000 mg/L,or were directly inoculated on the rooting medium above added IBA 1 mg/L ,which was better for rooting.
