摘要
目的 研究日本血吸虫 (中国大陆株 )脂肪酸结合蛋白 (Sj FABPc)重组抗原作为血吸虫病疫苗候选分子的潜能 ,并探讨Sj FABPc/Sj2 6GST融合蛋白作为复合疫苗的可能性。 方法 用亲和层析法制备Sj FABPc重组抗原和Sj FABPc/Sj2 6GST融合蛋白。将两种蛋白分别免疫BALB/c小鼠后进行攻击感染试验。结果 Sj FABPc及Sj FABPc/Sj2 6GST分别诱导小鼠产生了 2 3 6 0 % (P <0 0 5 )和 2 1 72 % (P >0 0 5 )的成虫减虫率 ,5 9 36 % (P <0 0 0 1)和 4 9 6 8% (P <0 0 0 1)的总减卵率。
Aim To evaluate the immunoprotective effect of Schistosoma japonicum fatty acid binding protein(rSj FABPc) and Sj FABPc/Sj26GST fusion protein Methods The Sj FABPc/Sj26GST fusion protein was prepared by affinity chromatography using glutathione Sepharose 4B The purified rSj FABPc was cleaved easily from the fusion with PreScissionTM Protease BALB/c mice immunized with rSj FABPc and Sj-FABPc/Sj26GST separately were each challenged with 40±1 S japonicum cercariae Results In BALB/c mine,the rSj FABPc and Sj FABPc/Sj26GST could induce 23 6%(P<0 05) and 21 72%(P>0 05) worm reduction,respectively,as well as 59 36%(P<0 001) and 49 68%(P<0 001) total egg reduction Conclusion rSj FABPc is partially effective against S japonicum
