摘要 目的 通过改进胰岛的制备方法 ,以提高胰岛的获取数量和质量 ,从而解决可供移植的有功能的胰岛数量不足这一矛盾。方法 在经典的胰岛制备方法基础上对其加以改进 ,即在胶原酶V分离液及Fio11-40 0纯化液中加入STI与BSA -FractionV后 ,进行胰岛的分离与纯化。结果 胰岛的产量为 (1,36 0± 2 75 )EIN/胰腺 (n =8) ,纯度为 85 % ,存活率为 90 % ,胰岛的生物学活性良好。 Objective To increase yield and quality of islets for transplantation by improving the technique of islets preparation. Methods The technique of islet preparation was improved on the base of classical methods, soybean typsin inhibitor and BSA-Fraction V were added into separation solution of collagenase V and the purified rate was 85% and survival rate was 90% by the modified method. Results The yield of islet was (1.360±275) EEIN per pancreas (n=8), and the purified rate was 85% and survival rate was 90% by the modified method, islet has better biological activity. Conclusion Improved method could obviously increase yield and quality of islets.
