摘要 〔目的〕介绍测定尿8-羟基脱氧鸟苷(8-OH-dG)含量的ELISA方法。〔方法〕利用8-OH-dG特异性单克隆抗体试剂盒,结合酶标仪进行定量测定。〔结果〕尿样测定值的批间和批间相对标准偏差分别为5.8l%~9 35%和6.42%~10.11%;同一个体一天中不同时点尿样的8-OH~dG含量之间没有显著性差异;80例健康志愿者的尿8-OH-dG含量为14,15±9.00ng/mgCr。其中男性为11.85±5.72ng/mgCr,女性为16.24±10.83ng/mgCr,女性的尿8-OH-dG含量明显高于男性;健康男性和女性尿8-OH-dG含量的95%参考值范围分别为0~25.77ng/mgCr和0~44.89ng/mgCr。〔结论〕应用ELISA试剂盒法可以简单灵敏、准确地测定尿8-OH-dG含量。 (Objective] A method for determination of urine 8 - hydroxyl - deoxyguanosine (8 - OH - dG) by ELISA was introduced. [Methods] Based on the specific monoclonal antibody, quantitative assay of urine 8 - OH - dG concentrations was conducted with the use of ELBA kit. [Results] The with - run RSD of urine samples ranged, bewtter 5.81% and 9.35% and between - run were 6.42% and 10.11%, respectively. There were no significant variations in the same persons among ruine samples at different times of one day. The average urine concentrations of 8 - OH - dGamong 80 healthy volunteers were 14.15±9. 00ng/mgCr, while 11.85±5.72ng/mgCr in males and 16.24 ±10. 83ng/mgCr in females. The urine 8 - OH - dG concentrations were higher in females than in males. The 95% reference ranges of urine 8 - OH - dG concentrations for healthy persons were 0 to 25.77 ng/mgCr and 0 to 44.89ng/ mgCr in males and females, respectively. [Conclusions] The method was simple, sensitive and accurate for analysis of urine 8 - OH -dG concentrations.
