摘要
铁是机体必需元素之一,同时又参与机体活性氧介质的生成。本文探讨了铁对过氧化氢诱导细胞凋亡的影响。方法是以细胞系K562为对象,采用流式细胞仪、DNA琼脂糖凝胶电泳和形态学的方法检测细胞凋亡的发生和细胞周期的改变。结果显示,0.001μM的H2O2作用K562细胞24小时,能明显地诱导细胞发生凋亡,作用持续至48小时;1mg/ml的FeSO4单独作用对细胞的增殖没有明显作用,也不诱导细胞凋亡;同时加入H2O2和FeSO4作用48小时,细胞凋亡比例明显低于H2O2作用组(P<0.01),并伴有细胞周期G2/M期的阻滞。结论:在本文培养体系中,铁不能诱导K562细胞凋亡,相反还对过氧化氢诱导的细胞凋亡具有保护作用,其作用机制可能是延长了细胞在G2/M期阻滞的时间,以利于受损细胞恢复,从而降低了凋亡比例。
Iron which is essential for cell growth and viability, participates in the formation of reactive oxygen species. To investigate the effect of iron on the apoptosis induced by hydrogen peroxide (H2 O2) , apopto-sis and the change of cell cycle of K562 cell were assessed by flow cytometry, agarose gel electrophoresis and microscopy. Results showed that cell apoptosis were induced by 0.001 uM H202 during 24-48 hours, but not by 1 mg/ml FeSO4 alone. Combined with H2 O2 and FeSO4 during 48 hours, the percentage of apoptosis was lowered than that by H2O2 alone, G2/M arrest was detected at the same time. Therefore, these findings showed that iron exposure could not induce cell apoptosis,in K562 cell culture, but inhibit apoptosis induced by H2O2. The mechanism may be to prolong the time of G2/M arrest to recover cells.
