摘要
目的 探讨全反式维甲酸 (ATRA)和苯乙酸钠 (Na-PA)两者对人肝癌细胞联合作用 .方法 应用台盼蓝染色、MTT法、流式细胞仪、甲胎蛋白测定等方法检测 ATRA和Na PA两者联合对人肝癌细胞的诱导分化作用 .结果 10μmol· L- 1 ATRA,2 .5 mmol· L- 1 Na PA及两者联合作用 ,可抑制人肝癌细胞株 SMMC- 772 1细胞的生长 ,使细胞恶性表型向正常转化 ,其联合作用组 7d抑制率达 6 6 .5 % ,并能明显降低甲胎蛋白的分泌量 4 9.2 % ,使细胞周期 G0 / G1 期延长5 8.7% ,S期下降 35 .8% (P<0 .0 5 ) ,影响 DNA合成 ,最终产生抑制增殖诱导细胞分化的作用 .结论 ATRA,Na PA及两者联合应用均可抑制 SMMC- 772 1细胞生长 ,诱导细胞分化 ,ATRA和 Na PA联合对人肝癌细胞的诱导分化有协同作用 .
AIM To investigate the effects of all trans retinoic acid in combination with phenylacete natrium on human hepatocellular carcinoma (HCC) cell line SMMC 7721. METHODS HCC cell line was exposed to all trans retinoic acid and phenylacete natrium separately and both agents in combining manner respectively. The morphologic and functional changes of the cell line were observed with benzo blue, MTT, flow cytometric analysis, image analysis and AFPmeasurement. RESULTS Single all trans retinoic acid or phenylacete natrium could inhibite the proliferation of human HCC cell line (SMMC 7721), which had obvious dose and time effect. After SMMC 7721 cells were treated with combined treatment of both agents for 7 d, the suppression rate was about 66.5%. The malignant phenotype of the cell line gradually disappeared while characteristics of mature cell were progressively increased. The secretory volume of AFP was reduced by 49.2%. The cells at G 0/G 1 phase were relatively increased by 58.7% and synthesis of DNA inhibited by 35.8% ( P <0.05). CONCLUSION The combined use of the two agents can inhibit proliferation and induce differentiation in the HCC cell more significantly than any other agent.
