摘要
目的:探讨型糖尿病患者血中单个核细胞内2Ca2+浓度变化及其与胰岛素受体活性变化的关系。TPK方法:荧光分光分析法测定例型糖尿病及例正常对照单个核细胞内40220Ca2+浓度,并用酶联免疫法测定单个核细胞胰岛素受体酪氨酸激酶()活性。TPK结果:型糖尿病患者单个核细胞内2Ca2+浓度显著高于正常对照组,胰岛素受体活性显著低于正常对照TPK组。相关分析显示型糖尿病患者单个核细胞内2Ca2+浓度与、显著正相关,与受体活性显著负相关。FBGFINSTPK结论:细胞内Ca2+浓度升高可能通过影响胰岛素受体活性参与胰岛素抵抗的发生。
Objective:To investigate the change of [Ca2+]i in type 2 diabetes mellites patients and its relationship with insulin receptor tyrosine kinase activity. Methods: The [Ca2+]i was assayed with fluorescence spectroscopic analysis and the insulin receptor tyrosine kinase activity was assayed with enzyme-linked immunosorbent assay method in circulating mononuclear cells in 40 type 2 diabetes mellites patients and 20 normal subjects. Results:The [Ca2+]i of circulating mononuclear cells in type 2 diabetes mellites patients was significantly higher than that in normal controls. The insulin receptor tyrosine kinase activity was significantly decreased in patients. The [Ca2+]i of circulating mononuclear cells in type 2 diabetes mellites patients was positively correlated with FBG, FINS, and negatively with the insulin receptor tyrosine kinase activity. Conclusion: The results suggest that elevation of [Ca2+]i may affect the insulin receptor tyrosine kinase activity and thus contributes to the insulin resistance.
