Lipopolysaccharide Intranigral Injection Reduces Dopamine Content in Nigrostriatal System of Rats

摘要 目的 :探讨黑质内注射脂多糖对黑质纹状体系统单胺类递质的影响。方法 :将 2 0只SD大鼠随机分成A、B、C 3组和对照组D ,取各组大鼠纹状体和黑质组织 ,采用高压液相色谱 电化学法 (HPLC ECD)检测该部位的单胺类递质含量。结果 :A、B、C 3组纹状体、黑质部位多巴胺及其代谢产物含量明显并持续降低 ,较D组均有显著性差异 ,但纹状体、黑质部位 5 羟色胺含量仅有一过性变化。结论 :黑质内注射脂多糖能使纹状体。 Aim:To investigate the effect of intranigral injection of the immunostimulant lipoplysaccharide(LPS) on monoamine transmitters in the nigrostriatal system.Methods:Twenty rats were randomly divided into 4 groups:group A(14 days after LPS injection),group B(21 days after LPS injection),group C(28 days after LPS injection)and group D(14 days after PBS injection).Animals were decapitated at different time and the monoamine transmitters in the striatum and substantia nigra were measured by high performance liquid chromatography with electrical detection(HPLC ECD).Results:In group A,B and C,the levels of dopamine and its metabolite(DOPAC) in the striatum and substantia nigra were significantly decreased,compared with group D.The content of serotonin in the striatum and substantia nigra showed transient change.Conclusion:Intranigral injection of LPS can significantly reduce the dopamine transmitter in the nigrostriatal system
