摘要
目的 :研究肝细胞肝癌和正常肝细胞间隙连接蛋白connexin 32 (Cx32 )、connexin 4 3(Cx4 3)的表达 ,及其对间隙连接通讯功能 (GJIC)的影响。方法 :应用细胞培养及流式细胞分析技术 (FCM) ,研究肝癌细胞系HHCC、SMMC 772 1和正常肝细胞系QZG细胞中Cx32和Cx4 3的表达。结合LuciferYellow划痕标记荧光传输技术 (SLDT) ,检测上述细胞的间隙连接通讯功能。结果 :流式细胞仪分析证实 ,Cx32蛋白在肝癌细胞系HHCC、SMMC 772 1和正常肝细胞系QZG细胞中表达的阳性率分别为 1 9%、0 7%和 99.0 % ;Cx4 3蛋白在HHCC、SMMC 772 1和QZG细胞中表达的阳性率分别为 7 3%、2 6 5 %和 99 1%。SLDT检测发现肝癌细胞HHCC ,SMMC - 772 1的间隙连接通讯功能较正常肝细胞QZG明显减弱。结论 :Cx32、Cx4 3蛋白在正常肝细胞中具有较高水平的表达 ,在肝癌细胞中表达水平显著降低 ;肝癌细胞的间隙连接通讯功能较正常肝细胞亦明显减弱。Cx32、Cx4 3表达调控异常引起的间隙连接通讯障碍可能与肝癌的发生密切相关。
Purpose To investigate expressions of gap junctional proteins connexin32(Cx32)and connexin43(Cx43) in human hepatocellular carcinoma cell lines and effects on gap junctional intercellular communication(GJIC). Methods Expressions of Cx32 and Cx43 were analyzed in human hepatocellular carcinoma cell lines HHCC, SMMC 7721 and normal liver cell line QZG using flow cytometry (FCM). Diffusion of lucifer yellow fluorescent probes among adjacent cells in HHCC, SMMC 7721 and QZG cell lines were carried out by scrape loading and dye transfer technique (SLDT) to reveal gap junctional intercellular communication in various cell lines. Results The positive percents of Cx32 were 0 7% in HHCC, 1 9% in SMMC 7721 and 99 0% in QZG; the positive percents of Cx43 were 7 3% in HHCC, 26 5% in SMMC 7721 and 99 1% in QZG. SLDT showed that QZG cells had better gap junctional intercellular communication than HHCC and SMMC 7721 cells. Conclusion The results indicate that a dramatic decrease in gap junctional intercellular communication due to abnormal expressions of Cx32 and Cx43 might contribute to carcinogenesis of human hepatocellular carcinoma.
