摘要
应用PHA—LCM刺激的甲基纤维素体系进行了25例ANLL骨髓细胞去T细胞后的CFU-L培养及Har和Adr药物敏感试验。发现CFU-L体外对Har敏感性与用Har治疗的病人疗效相关,表明Har体外药敏试验能提示临床疗效、CFU-L对Adr的敏感性与Har具有高度一致性。
With PHA-LCM stimulated methylcellulose system, the culture results of bone marrow cells deprived of T-lymphocyte, of 25 patients with ANLLwere studede in this article.The morphology and ability of self-renewal of the colony cells suggested that the colonies were originated from leukemic progenitor cells (CFU-L) .The number of colony growth was linearly correlated with that of cells plated.It was found that the colony production rate was much higher in type M5 than that in M1-3 according to the FAB classification.The sensitivity of CFU-L to harringtonine (Har) in vitro was well correlated with the results of clinical treatment when the drug concentration was 0.5ug/ ml.This result suggested that the in vitro sensitivity to Har could be used as a predictive index for ANLL patients treated with Har.We also found that the in vitro sensitivity of CFU-L to adrimycin was very similar to that of Har.
