摘要
目的 :采用高效液相色谱法测定黄连上清丸中黄芩苷的含量 ,以控制该制剂的质量。方法 :以 C1 8化学键合硅胶柱分离黄芩苷 ,以甲醇 -水 -醋酸 (40∶ 6 0∶ 1)为流动相 ,U V检测波长 2 74nm进行测定。结果 :黄芩苷峰与其它组分峰能够基线分离 ,理论板数以黄芩苷峰计算为 35 0 0 ;方法的平均加样回收率为 10 0 .8% (n =5 ) ;5次独立测定的相对偏差为 RSD =0 .92 % ,黄芩苷在 0 .2 5~ 2 .5μg范围内 ,进样量与吸收面积值呈良好的线性关系。结论 :本法测定黄连上清丸中黄芩苷的含量 ,结果准确、重复性好。
Objective: To control the quality of Huanglianshangqing pills through determination of baicaline content in Huanglianshangqing pills by Reversed Phase HPLC. Method: Baicaline was separated on spherisorb C 18 column by using methanol acetic acid water(40∶ 1∶ 60)as mobile phase and UV detection wavelength at 274 nm. Results: The number of theoretica1 plates calculated for baicaline peak was 3500; The average recovery of standard addition was 98.8%(n=5). RSD got by 5 time of determination was 0.92%. There was a good linear relationship between the concentration of Baicaline and absorption area value in the range of 0.25~2.5 μg. Conclusion: The method was highly reproducible and accurate.
