摘要 目的 探讨 Vit.C,E对 6 0 Coγ线照射家兔的保护作用以及剂量效应关系 .方法 中国本兔随机分为 12组 ,每组5只 .Vit.C 5组 iv,剂量分别为 10 ,2 0 ,4 0 ,80和 16 0 mg·kg- 1 ,Vit.E油剂 ig5组 ,剂量分别为 5 ,10 ,2 0 ,4 0和 80 mg·kg- 1 .第 3日进行 6 0 Coγ线 (4.5 Gy)照射家兔 .第 5日处死、剖腹取肝脏和腹腔静脉血 ,分别测定超氧化物歧化酶 (SOD)的催化活性和丙二醛 (MDA)的含量变化 .结果 辐射动物补充 Vit.C和 Vit.E组的肝 SOD的催化活性与阴性对照组相比明显升高 (P<0 .0 1) ,Vit.E的作用更明显 ,呈现了一定剂量效应关系 ;各 Vit.C组肝脏 MDA的含量变化不显著 (P>0 .0 1) ,但血清 MDA的含量轻微下降 ;各 Vit.E组肝脏和血清中的 MDA的含量均显著降低 (P<0 .0 1) ,且呈现出一定的剂量效应关系 .结论 单独使用 Vit.C的抗氧化作用 ,有时还会加重损伤 ;单独使用 Vit.E具有一定的 SOD活性保护和MDA清除作用 ,存在剂量效应关系 。 AIM To study the dose effects relationship between Vit.C, Vit.E, and lipid peroxidation through the model of rabbit injured by radiation. METHODS Chinese rabbits were chosen as the experimental animals and were randomly divided into twelve groups which each contained five. They were negative control group, positive group, and ten trial groups of Vit.C, Vit.E. The trial groups contained five different dosage of Vit.C and five different dosage of Vit.E. Peanut oil was selected as diluent which had been getting rid of Vit.E. The five groups were supplied by injection of vein with the dosages of Vit.C 10, 20, 40, 80 and 160 mg·kg -1 every day respectively. The five Vit.E groups were supplied by gavages with the dosage of Vit.E 5, 10, 20, 40 and 80 mg· kg -1 , every day espectively too. On the third day all groups, except negative control group, were radiated by 60 Co γ ray with the dose of 4.5 Gy. Then they were fed as before, and on the sixth day all rabbits were sacrificed. Their blood and livers were removed for the analysis of changes of catalytic activity of SOD and content of MDA. RESULTS After the radiation, the catalytic activities of hepatic SOD in the radiation groups were higher than those in the negative control group ( P <0.01), and the trial groups of Vit.C and Vit.E showed some protective effects on rabbit against radiation, and there was a dose effect relationship between the Vit.E and radiation injury. The content of hepatic MDA in Vit.C group was higher than positive group except Vit.C 80 mg·kg -1 group ( P <0.01). Vit.C showed some protective effects against the increase of MDA. The markable inhibition of Vit.E on hepatic and serum MDA emerged ( P <0.01), and these showed the dose effect relationship between the dosage of vit.E and protection against injury of irradiation. CONCLUSION Vit.C is limited in antioxidation, sometimes can enhance injury; Vit.E is effective against free radical injury; Vit.E has protective action on SOD catalytic activity and inhibition on MDA, which shows dose effect relationship. Vit.E cannot protect against radiation of peripheral blood and immunological cells.
