摘要
目的 :探讨受体相关蛋白的 (RAP)在肾皮质的表达与脂代谢的关系。方法 :应用编码 319氨基酸的RAP基因片段克隆表达的融合蛋白 ,制备针对该受体相关蛋白的抗RAP抗体 ,用免疫印迹杂交方法及免疫组化法分析家兔高胆固醇血症模型的肾皮质中RAP的表达情况。结果 :模型组与对照组在 4 4KDa处均有RAP杂交带 ,RAP在高胆固醇血症模型肾皮质中的表达量明显高于对照组。免疫组化显示RAP主要分布于肾小球毛细血管基底膜、肾小管周围间质内。结论
Objective: The structure of the RAP (receptor-associated protein) is similar to the form of receptor of low density lipoprotein. The RAP belonged to the members of LDL-R family. In order to research the relationship between the RAP and metabolism of lipid, expression and distribution of RAP in the model of hypercholesterolemia in kidney was detected. Methods:The model of hypercholesterolemia was made. The GST-RAP fusion protein, which was constructed by inserting the cDNA encoding the (RAP) of 319 amino acid into the PGEX, was induced by IPTG. The antibody against the RAP was inducted by immunizing the rabbit with the GST-RAP fusion protein. Expression and distribution of RAP in kidney in the model and control was detected by the antibody with Western-bolt and immunohistochemistry. Results:Western-blot analysis showed that expression of RAP in 44KDa band in kidney was higher in the model than that in the control. Immunohistochemistry result showed that distribution of RAP in kidney lay in glomerular base membrane and in medium around the proximal convoluted. Conclusion: The RAP may play a role in the process of metabolism of lipid.
