摘要
从某猪场仔猪体内分离到一株 PPRSV毒株 ,该毒株能在 Marc-1 4 5细胞上增殖产生致细胞病变 ,该病变能被 PPRSV美洲型阳性血清所抑制 ,不被乙脑病毒、猪细小病毒、伪狂犬病毒阳性血清所抑制 ,经美洲型 PPRSV荧光抗体染色呈阳性反应 ,电镜观察到直径 55~ 60 nm的病毒粒子 ,接种阴性仔猪未见临床症状异常 ,但抗体检测阳性。命名该分离毒为 PPRSV-S3毒株。在 S3弱毒株分离鉴定的基础上 ,进一步在猪体上对其致病性和免疫力进行研究。S3毒株接种仔猪后 ,仔猪不表现任何症状 ,也不向外排毒。4~ 5周龄的仔猪接种 S3株后可产生坚强的免疫力 ,免疫后 4~ 6个月能抵抗强毒攻击。后备母猪配种前 2~ 3周免疫接种 S3株 ,怀孕 90~95d攻强毒 ,未发生流产、死胎、木乃伊胎、产弱仔等繁殖障碍现象。结果表明 ,S3是一株自然分离的弱毒株 ,且具有良好的安全性和免疫力。
Isolated a PPRSV strain from internal organs of fetus. The strain can increase and conduce CPE on Marc 145 cell. The CPE were restrained by positive serum of American genotype PPRSV, but JEV? PPV? PRV positive serum did not restrain it. The strain showed positive response through fluorescent antibody dyeing of American genotype PPRSV. Electron microscopic observed that diameter of virus particle was 55~60 nm . The strain inoculated negative piglets that did not show any clinical symptoms,the serological test showed positive. The strain was denominated PPRSV S3 strain. Furthermore ,the Safety and immunigenicity of S3 strain were studied. 4~5 weekly old baby pigs inoculated with S3 stainshowed protection against challenge exposure to virulent PRPSV, and didn`t show any clinical signs and secret virus as well. Replacement gilts were inoculated with S3 strain 2~3 weeks before mating,challenged with virulent PRRSV at 90~95 days after pregnancy .the pregnant swine didn`t showe any reproductive failure such as abortion,mummification of fetuses , stillbirth , birth of weak pigs and so on. The results indicated that S3 strain had good safety and immunigenicity , and was a natural attenuated PPRSV.
