摘要 目的 介绍实验性变态反应性脑脊髓炎 (EAE)的致敏抗原———髓鞘素的一种制备方法。方法 取牛脊髓白质加入蔗糖溶液制成脊髓匀浆 ,在 4℃通过不连续梯度离心 ,得到粗制的髓鞘素 ,进一步去除其中的磷脂、胆固醇和核酸等成分 ,得到纯化的髓鞘素。计算其含量 ,检测其主要成分及相对分子质量 ,并通过对豚鼠进行免疫 ,检测其诱发EAE的活性。结果 所提取的髓鞘素具有与文献报道结果类似的理化特性和强大的EAE诱发活性。结论 本方法提纯的髓鞘素有强的免疫活性 ,易取材 ,操作简便 ,便于推广。 Objective To introduce a method for extraction and identification of myelin:a sensitization antigen to experimental autoimmune encephalomyelitis.Methods After discontinuous gradient centrifugation, crude myelin was obtained from bovine spinal cord white matter.By means of removing phosphoglycerides, cholesterol and nucleic acid, purified myelin was obtained, and was identified by calculating its purity, main compositions and relative molecule weight.Additionally, its immunologic activity was observed by inducing EAE in guinea pigs.Results The extracted myelin has chemical characteristics and biological activity, and successfully induced experimental autoimmune encephalomyelitis (EAE).Conclusion The extracted myelin has strong immunologic activity, the advantages of the method being convenient materials, easy to manipulate and suitable to wide application.
