摘要 目的 介绍一套高效率的大鼠胎心细胞的分离培养方法和应注意的事项。方法 无菌条件下取胎心 ,经酶解消化法分离胎心细胞 ,通过差速贴壁和添加Brdu来提高胎心细胞纯度。结果 培养的心肌细胞的纯度可达94% ,活细胞数 >85 % ,形态典型 ,可同步自律搏动达 2~ 3周。结论 该方法可为实验研究提供纯度高、活细胞数较多。 Objective\ To introduce a high efficient method of rat fetal cardiomyocyte isolation and culture. Methods\ Cardiomyocytes were harvested from the hearts of fetal rats sterilely and were isolated by enzymatic digestion.To improve the purity of the cardiomyocytes Brdu was added and preplating method was used.Results\ The purity of the cultured cardiomyocytes can reach 94%.The number of cardiomyocytes alive is more than 85%.The cells can contract synchronously and spontaneously for 2~3 weeks.Conclusion\ This method can offer more pure, more active and more typical fetal cardiomyocytes for further study.
