摘要
为了从荷叶中提取并制备高纯度的原花青素,采用大孔树脂和高速逆流色谱技术(HSCCC)分离纯化荷叶中的原花青素。以正己烷∶乙酸乙酯∶水=1∶80∶80的溶剂体系,得到三种目标组分G1、G2、G3,经高效液相色谱分析其组成可知,三种组分对应四种物质分别为儿茶素、表儿茶素、表儿茶素没食子酸酯和原花青素B2,并对三种组分进行抗氧化活性研究。结果表明,G1、G2和G3对DPPH自由基的半数清除率(IC50)分别为0.12 mg/L、0.08 mg/L、0.09 mg/L;对ABTS^+自由基的半数清除率(IC50)分别为0.48 mg/mL、0.28 mg/mL、0.21 mg/mL;对羟基自由基的半数清除率(IC50)分别为0.29 mg/mL、0.35 mg/mL、0.39 mg/mL,证实了这三种组分都具有较好的抗氧化活性。
In order to extract high-purity proanthocyanidins from lotus leaves,macroporous resin and high-speed countercurrent chromatography(HSCCC)were used to separate and purify proanthocyanidins from lotus leaves.The solvent components of n-hexane∶ethyl acetate∶water=1∶80∶80 were used to obtain three target components G1,G2 and G3.The composition was analyzed by high performance liquid chromatography.The analysis indicated that the three components corresponded to four substances:catechin,epicatechin,epicatechin gallate and proanthocyanidin B2 The antioxidant activities of the three components were studied.The results showed that the half-clearance rate(IC50)of DPPH radicals was 0.12 mg/L,0.08 mg/L,0.09 mg/L,and the half-clearance rate(IC50)of ABTS+radicals was 0.48 mg/mL,0.28 mg/mL,0.21 mg/mL,respectively.The half-resolution rate(IC50)of hydroxyl radicals was 0.29 mg/mL,0.35 mg/mL,and 0.39 mg/mL,respectively.All three components isolated have good antioxidant activity.
